摘 要:目的评价凉粉草多糖(MCP)对于葡聚糖硫酸钠(DSS)诱导的小鼠溃疡性结肠炎(UC)的影响。方法采用碱水提取、醇沉等方法制备MCP,测定其多糖含量、蛋白质含量及单糖组成,通过凝胶渗透色谱(GPC)测定MCP的分子量。建立DSS诱导UC小鼠模型,将小鼠分为空白组(Control)、模型组(Model,3%DSS)、阳性药组(5-ASA,100 mg/kg)、低剂量组(MCP,100 mg/kg)和高剂量组(MCP,200 mg/kg)连续灌胃给药10 d,考察MCP对UC小鼠体质量和疾病活动指数、结肠肠段长度、结肠病理切片、结肠上皮细胞生长情况、血液炎性细胞数量、结肠组织中IL-6和TNF-α水平以及结肠上皮细胞F4/80和CD206的表达影响。结果MCP中多糖含量为34.56%、蛋白质含量为5.65%,单糖组成为甘露糖(Man)、鼠李糖(Rha)、半乳糖醛酸(GalA)、葡萄糖(Glc)、半乳糖(Gal)、木糖(Xyl)和阿拉伯糖(Ara)等7种单糖,分子量为14753 Da。MCP能够显著降低UC小鼠血液炎性细胞数量,降低UC小鼠结肠组织中炎症因子TNF-α和IL-6水平,降低结肠巨噬细胞的表达,从而减轻结肠炎小鼠炎症,MCP能够促进小鼠结肠巨噬细胞M2极化发挥抗炎作用。结论MCP可能通过抑制DSS诱导的UC小鼠的炎症发挥抗UC作用。Objective To evaluate the effect of Mesona chinensis total polysaccharides(MCP)on dextran sodium sulfate(DSS)-induced ulcerative colitis(UC)in mice.Methods MCP was prepared by alkaline water extraction and alcohol precipitation,and its polysaccharide content,protein content and monosaccharide composition were determined,and the molecular weight of MCP was determined by gel permeation chromatography(GPC).A mouse model of DSS-induced UC was established,and the mice were divided into blank group(Control),model group(3%DSS),positive drug group(5-ASA,100 mg/kg),low-dose group(MCP,100 mg/kg)and high-dose group(MCP,200 mg/kg)for 10 days.Effects of colonic epithelial cell growth,number of inflammatory cells in blood,IL-6 and TNF-αlevels in colonic tissues,and expression of F4/80 and CD206 in colonic epithelial cells.Results The content of MCP polysaccharides was 34.56%,the protein content was 5.65%,and the monosaccharide composition was 7 monosaccharides,including mannose(Man),rhamnose(Rha),galacturonic acid(GalA),glucose(Glc),galactose(Gal),xylose(Xyl)and arabinose(Ara),with a molecular weight of 14753 Da.MCP could significantly reduce the number of inflammatory cells in the blood of UC mice,reduce the levels of inflammatory factors TNF-αand IL-6 in the colon tissue of UC mice,and reduce the expression of colon macrophages,thereby reducing inflammation in colitis mice.Conclusion MCP may play an anti-UC role by inhibiting DSS-induced inflammation in UC mice.
