摘  要:目的探讨miRNA-150在宫颈癌中表达的临床意义及对宫颈癌细胞上皮间质转化的作用机制。方法收集2022年6月-2023年6月我院妇产科收治的120例接受宫颈癌根治手术患者的癌组织及相对应的癌旁组织标本,RT-PCR检测组织中miR-150相对表达量,探究miR-150表达和临床比例特征相关性;RT-PCR检测宫颈癌和正常宫颈细胞中miR-150相对表达量,将Hela细胞随机分为miR-NC组、miR-150组、anti-miR-150组、anti-miR-NC组。采用CCK-8、Transwell法、流式细胞仪分别检测细胞增殖、侵袭和凋亡能力;蛋白质印迹法检测细胞中E-Cadherin、N-Cadherin、Vimentin、PDCD4蛋白表达;生物信息学预测miR-150的靶基因,双荧光素酶报告基因检测miR-150和FOXO4的靶向关系。结果与癌旁组织(1.00±0.10)相比,宫颈癌组织中miR-150相对量(1.96±0.24)表达明显升高(P<0.01);miR-150高表达和FIGO分期、淋巴结转移和分化程度有显著相关性(P<0.05)。和Ect/E6E7细胞(1.00±0.10)相比,宫颈癌Hela、SiHa和CasKi细胞中miR-150相对表达量(分别为2.36±0.35)、1.89±0.24、1.45±0.19)均明显升高(P<0.05)。与miR-NC组相比,miR-150组48 h(1.26±0.13)、72 h(0.62±0.08)时Hela细胞增殖能力,侵袭数目(261.09±30.26)、细胞中N-Cadherin(1.62±0.28)和Vimentin蛋白(1.43±0.16)表达明显增加,凋亡率(1.02±0.23)及细胞中E-Cadherin蛋白(0.12±0.02)表达明显降低(P<0.05);与anti-miR-NC组相比,anti-miR-150组48 h(1.58±0.20)、72h(0.80±0.10)时Hela细胞增殖能力,侵袭数目(67.24±10.13)、细胞中N-Cadherin(0.31±0.05)和Vimentin蛋白(0.24±0.05)表达明显降低,细胞凋亡率(15.84±1.26)及细胞中E-Cadherin蛋白(0.98±0.13)表达明显增加(P<0.05)。与miR-NC组相比,转染野生型FOXO4(FOXO4-WT)时miR-150组荧光素酶活性(0.31±0.06)和细胞中FOXO4蛋白(0.11±0.02)表达明显降低(P<0.01)。结论miR-150在宫颈癌组织中高表达,与FIGO分期、淋巴结转移和分化程度有显著相关性;过表达miR-150可促进�Objective To investigate the clinical significance of miRNA⁃150 expression in cervical cancer and its mechanism of action on epithelial mesenchymal transition of cervical cancer cells.Methods 120 cancer tis⁃sues and corresponding adjacent tissue samples from patients undergoing radical cervical cancer surgery in our hospi⁃tal's obstetrics and gynecology department from June 2022 to June 2023 were collected.The relative expression of miR⁃150 in the tissues was detected by RT⁃PCR to explore the correlation between miR⁃150 expression and clinical proportional characteristics;RT⁃PCR was used to detect the relative expression of miR⁃150 in cervical cancer and normal cervical cells.Hela cells were randomly divided into miR⁃NC group,miR⁃150 group,anti miR⁃150 group,and anti miR⁃NC group.Cell proliferation,invasion,and apoptosis were detected using CCK⁃8,Transwell method,and flow cytometry,respectively;Protein blotting was used to detect the expression of E⁃Cadherin,N⁃Cadherin,Vi⁃mentin,and PDCD4 proteins in cells;Bioinformatics predicts the target genes of miR⁃150,and dual luciferase re⁃porter genes detect the targeting relationship between miR⁃150 and FOXO4.Results Compared with the adjacent tissues(1.00±0.10),the relative expression of miR⁃150(1.96±0.24)in cervical cancer tissue was significantly in⁃creased(P<0.01);The high expression of miR⁃150 was significantly correlated with FIGO staging,lymph node me⁃tastasis,and differentiation(P<0.05).Compared with Ect/E6E7 cells(1.00±0.10),the relative expression of miR⁃150 in cervical cancer Hela(2.36±0.35),SiHa(1.89±0.24),and CasKi(1.45±0.19)cells was significantly in⁃creased(P<0.05).Compared with the miR⁃NC group,the miR⁃150 group significantly increased Hela cell prolifera⁃tion ability,invasion number(261.09±30.26),N⁃Cadherin(1.62±0.28),and Vimentin protein(1.43±0.16)ex⁃pression at 48 hours(1.26±0.13)and 72 hours(0.62±0.08),while the apoptosis rate(1.02±0.23)and E⁃Cadherin protein(0.12±0.02)expressi
