摘 要:糖胺聚糖具备低毒副作用以及高生物活性,在抗肿瘤、抗血栓、降低血糖和免疫调节等方面具有生理活性。蜚蠊的生活能力很强,容易获取,并且作为中药已用于中医治疗疾病已有上千年历史,本文对糖胺聚糖提取工艺进行优化,采用化学酶法联合丙酮循环脱脂工艺,通过乙醇沉淀的方式分别从蜚蠊虫体、蜚蠊腿部提取糖胺聚糖,借助咔唑反应对蜚蠊虫体、蜚蠊腿部的糖胺聚糖进行定量分析,结果表明,蜚蠊虫体、蜚蠊腿部的糖胺聚糖提取率分别达到0.743%和0.962%,二糖组分分析得出,蜚蠊二糖以△UA2S-GlcNAc4S6S组成为主,还存在少量的△UA2S-GlcNAc6S。生物活性分析显示,从蜚蠊各组织中提取的糖胺聚糖均具有较高的抗氧化活性。其中,质量浓度为10 mg/mL的蜚蠊虫体糖胺聚糖的总抗氧化能力(T-AOC)最高,为0.8704μmol/mL,但因样本浓度不足,其DPPH·自由基清除率为负值。本研究为制备分析蜚蠊来源的药用糖胺聚糖活性寡糖提供了研究基础。Glycosaminoglycan has low toxic side effects and high biological activity,and has physiological activities such as anti-tumor,anti-thrombosis,lowering blood sugar and immune regulation.Cockroaches have strong living ability and are easy to obtain.As an important part,it has been applied in the treatment of diseases in traditional Chinese medicine for thousands of years.The glycosaminoglycan was extracted from the cockroach body and legs by ethanol precipitation,and the glycosaminoglycan of the cockroach body and legs was quantitatively analyzed by carbazole reaction.The results showed that the extraction rate of glycosaminoglycan of the cockroach body and legs was 0.743%and 0.962%,respectively.The disaccharide component analysis showed that,the disaccharides were mainly△UA2S-GlcNAc4S6S,with a small amount of△UA2S-GlcNAc6S.Bioactivity analysis showed that the glycosaminoglycans extracted from various tissues of cockroaches had high antioxidant activity.The total antioxidant capacity(T-AOC)of 10 mg/mL snail slime glycosaminoglycans was the highest(0.8704μmol/mL),but the DPPH·radical scavenging rate was negative due to insufficient sample concentration.This study provided research basis for preparation and analysis of active oligosaccharides derived from medicinal glycosaminoglycans of cockroach.
