多波长HPLC法同时测定黄石感冒片中芦丁、阿魏酸、迷迭香酸和木犀草素的含量

摘　　要：目的:建立多波长HPLC法同时测定黄石感冒片中芦丁、阿魏酸、迷迭香酸和木犀草素4种成分的含量。方法:以芦丁、阿魏酸、迷迭香酸和木犀草素为对照品,采用Apollo C 18250 mm×4.6 mm,5μm色谱柱,流动相为乙腈-0.1%磷酸溶液梯度洗脱,流速为1.0 mL/min,柱温30℃,在不同检测波长(芦丁256 nm、阿魏酸322 nm、迷迭香酸328 nm、木犀草素348 nm)下,对4个指标成分进行定量分析。结果:芦丁在0.0910~0.9100μg范围内呈现良好的线性关系,相关系数R 2=0.9991;阿魏酸在0.0114~0.2276μg范围内呈现良好的线性关系,相关系数R 2=0.9996;迷迭香酸在0.0113~0.2266μg范围内呈现良好的线性关系,相关系数R 2=0.9992;木犀草素在0.0130~0.2606μg范围内呈现良好的线性关系,相关系数R 2=0.9996。芦丁平均回收率为100.72%(RSD=0.92%,n=6);阿魏酸平均回收率为94.71%(RSD=1.50%,n=6);迷迭香酸平均回收率为99.30%(RSD=1.16%,n=6);木犀草素平均回收率为99.20%(RSD=1.23%,n=6)。结论:本方法专属性强,准确度高,重复性及可行性好,可以用于黄石感冒片中芦丁、阿魏酸、迷迭香酸和木犀草素4种成分的含量测定。Objective To establish a multi wavelength HPLC method for simultaneous determination of rutin,ferulic acid,rosmarinic acid,and luteolin in Huangshi Ganmao tablets.Methods Using rutin,ferulic acid,rosmarinic acid,and luteolin as control substances,an Apollo C 18250 mm×4.6 mm,5μm chromatographic column was used.The mobile phase was a gradient elution of acetonitrile 0.1% phosphoric acid solution,with a flow rate of 1.0 mL/min and a column temperature of 30℃.The four indicator components were quantitatively analyzed at different detection wavelengths(rutin 256 nm,ferulic acid 322 nm,rosmarinic acid 328 nm,luteolin 348 nm).Results Rutin showed a good linear relationship in the range of 0.0910-0.9100μg,the correlation coefficient R 2=0.9991;Ferulic acid showed a good linear relationship in the range of 0.0114-0.2276μg with R 2=0.9996.Rosmarinic acid showed a good linear relationship in the range of 0.0113-0.2266μg with R 2=0.9992.Luteolin showed a good linear relationship in the range of 0.0130-0.2606μg with R 2=0.9996.The average recovery of rutin was 100.72%(RSD=0.92%,n=6).The average recovery of ferulic acid was 94.71%(RSD=1.50%,n=6).The average recovery of rosmarinic acid was 99.30%(RSD=1.16%,n=6).The average recovery of luteolin was 99.20%(RSD=1.23%,n=6).Conclusion The method showed high specificity,accuracy,good repeatability and feasibility.Meanwhile,this method can be used for the determination of rutin,ferulic acid,rosmarinic acid and luteolin in Huangshi Ganmao tablet.
