摘  要:β-半乳糖苷酶催化乳糖水解得到的低聚半乳糖(Galactooligosaccharides,GOS),具有维持肠道菌群稳定、免疫调节和抗炎等生物功能。文章通过酶活力以及转糖苷活力的测定,确定乳酸克鲁维酵母来源的β-半乳糖苷酶制备GOS。通过单因素试验得到不同的加酶量、反应时间、反应温度和反应pH对GOS产率有显著性影响,而不同的底物浓度影响不大。利用响应面试验优化GOS产率,获得该酶水解乳糖转化合成GOS的最佳工艺条件。结果表明:底物浓度200 mg/mL、加酶量0.030%、反应温度36.9℃、反应时间2.5 h、反应pH值6.7为最优条件。GOS产率可达(12.61±0.17)%。优化后的工艺参数可为GOS的定向合成提供理论依据。β-galactosidase catalyzed the hydrolysis of Galactooligosaccharides,which has the biological functions of maintaining intestinal flora stability,immune regulation and anti-inflammatory.In this study,β-galactosidase derived from Kluyveromyces lactate was determined by the determination of enzyme activity and transglycoside activity to prepare GOS.The results of single factor test showed that different enzyme dosage,reaction time,reaction temperature and reaction pH had significant effects on GOS yield,but different substrate concentration had little effect.On this basis,the response surface experiment was used to optimize the yield of GOS,and the optimum technological conditions for the synthesis of GOS were obtained.The results showed that the optimal conditions were substrate concentration of 200 mg/mL,enzyme dosage of 0.030%,reaction temperature of 36.9℃,reaction time of 2.5 h and reaction pH 6.7.GOS yield was(12.61±0.17)%.The optimized process parameters can provide a theoretical basis for the directional synthesis of GOS.
