摘 要:目的分析睡眠剥夺对大鼠焦虑样行为和海马、前额叶皮质抗氧化水平的影响。方法选取SD雄性大鼠12只,随机分为对照组(6只)和睡眠剥夺组(6只)。采用改良多平台水环境法建立72 h快速眼动睡眠剥夺模型,所有大鼠均进行高十字架迷宫实验(EPMT),检测大鼠焦虑样行为。通过蛋白印迹法检测大鼠海马、前额叶皮质超氧化物歧化酶2(SOD_(2))的水平。对比两组大鼠开放臂停留时间百分比(OT%)、闭合臂停留时间百分比(CT%)及海马、前额叶皮质组织SOD_(2)/β-肌动蛋白(β-actin)的表达情况。结果睡眠剥夺组高十字架迷宫实验中OT%(0.08490±0.06185)显著低于对照组的(0.19447±0.04429),CT%(0.91511±0.06186)显著高于对照组的(0.80547±0.04426)(P<0.05)。睡眠剥夺组海马、前额叶皮质组织β-actin的表达与对照组对比差异不具有显著性(P>0.05);睡眠剥夺组海马、前额叶皮质组织SOD_(2)/β-actin表达水平分别为(0.05008±0.01485)、(0.05465±0.01271),低于对照组的(0.06427±0.00354)、(0.06990±0.00756)(P<0.05)。结论72 h快速眼动睡眠剥夺可引起大鼠焦虑样行为,其机制可能与海马、前额叶皮质抗氧化水平降低有关。Objective To analyze the effect of sleep deprivation on anxiety-like behavior,antioxidant levels in the hippocampus,and prefrontal cortex in rats.Methods 12 SD male rats were randomly divided into control group(6 rats)and sleep deprivation group(6 rats).A 72 h REM sleep deprivation model was established by modified multi-platform water environment,and all SD rats were subjected to elevated plus-maze test(EPMT)to detect anxiety-like behavior.The level of superoxide dismutase 2(SOD_(2))in the hippocampus and prefrontal cortex was examined by western blot.The percentage time spent on open arms(OT%)and percentage time spent on closed arm(CT%)and the expression of SOD_(2)/β-actin in hippocampal and prefrontal cortex of rats in the two groups were compared.Results The OT%of(0.08490±0.06185)in the elevated plus-maze test of the sleep deprivation group was significantly lower than(0.19447±0.04429)of the control group,and the CT%of(0.91511±0.06186)was significantly higher than(0.80547±0.04426)of the control group(P<0.05).There was no significant difference in the expression ofβ-actin in hippocampus and prefrontal cortex between the sleep deprivation group and the control group(P>0.05).SOD_(2)/β-actin expression levels in hippocampus and prefrontal cortex of the sleep deprivation group were(0.05008±0.01485)and(0.05465±0.01271),which were lower than(0.06427±0.00354)and(0.06990±0.00756)of the control group(P<0.05).Conclusion 72 h REM sleep deprivation can cause anxiety-like behavior in rats,and the mechanism may be related to the reduced antioxidant level in the hippocampus and prefrontal cortex.
