摘　　要：目的:研究喉癌中第10号染色体缺失的磷酸酶及张力蛋白同源蛋白(PTEN)和人端粒酶逆转录酶(hTERT)在喉癌中的表达特点。方法:应用Western blot及qRT-PCR研究喉癌组织和正常组织中PTEN和hTERT表达的相关性,以及与喉癌临床病理特征的关系。结果:Western blot检测喉癌组织中hTERT蛋白的表达高于正常组织(P<0.001),PTEN蛋白在正常组织的表达高于喉癌组织(P<0.001)。qRT-PCR检测喉癌组织中hTERT mRNA的表达高于正常组织(P<0.001);PTEN mRNA在正常组织的表达高于喉癌组织(P<0.001)。喉癌组织中PTEN和hTERT的表达均呈负相关(P<0.001)。结论:喉癌中PTEN的表达可能对hTERT起到调控作用,hTERT有可能成为喉癌早期诊断的分子标志物。Objective:To investigate the expression characteristics of phosphatase and tensin homologue deleted on chromosome 10(PTEN)and human telomerase reverse transcriptase(hTERT)in laryngeal cancer.Methods:Western blot and qRT-PCR were used to analyze the correlation between hTERT and PTEN expression in the laryngeal cancer tissues,and the relationship of the two proteins with clinical pathological features of laryngeal cancer.Results:Western blot assay revealed that the expression of hTERT protein was significantly higher than that in normal tissues(P<0.001),and the expression of PTEN protein in normal tissues was markedly higher than that in laryngeal cancer tissues(P<0.001).qRT-PCR detection indicated that the expression level of hTERT mRNA in laryngeal cancer tissues was significantly higher than that in normal tissues(P<0.001),and the expression of PTEN mRNA in normal tissues was notably higher than that in the laryngeal cancer tissues(P<0.001).PTEN and hTERT expression was in negative correlation in the laryngeal cancer tissues(P<0.001).Conclusion:PTEN expression in laryngeal cancer plays a regulating role in hTERT,which suggests that hTERT may be a molecular marker for the early diagnosis of laryngeal cancer.
