摘 要:试验旨在探究易捕鲤生肌调节因子家族基因及养殖模式对易捕鲤生肌调节因子家族基因表达、生长和肌肉质构的影响。选取规格均一的易捕鲤90尾,随机分为2组,每组3个重复,每个重复15尾易捕鲤。静水组(CK组)和流水组(EG组)均投喂鲤鱼鱼种配合饲料。预试期1 w,正式试验期73 d。结果显示,通过NCBI BLAST比对发现易捕鲤肌肉生肌因子6 (myf6)、成肌分化因子(myoD)与鲤鱼高度同源,肌细胞生成素(myoG)在进化上明显处于一个单独的分支,使用β-actin作为内参基因,通过荧光实时定量PCR发现,在静水养殖模式下,易捕鲤myf6、myoD表达量显著高于流水组(P<0.05);在2种养殖模式下,myoG的表达无显著差异(P>0.05)。在2种养殖模式下,易捕鲤期的生长性能、肌肉质构和营养特性均无显著差异(P>0.05)。研究表明,受集约化养殖成本较高的影响,应慎重选择易捕鲤作为集约化循环水养殖对象。The aim of this experiment was to explore the myogenic regulatory factor family genes in easy-catching carp,and the effects of culture mode on gene expression,growth and muscle texture of Cyprinus carpio var.Yibu.A total of 90Cyprinus carpio var.Yibu of uniform size were selected and randomly divided into two groups,with three replicates per group and 15 carps per replicate.Both the still water group(CK group) and the flowing water group(EG group) were fed with carp fingerling compound feed.The pre-test period was one week,and the formal test period was 73 days.The results showed that through NCBI BLAST comparison,the myogenic factor(myf6) and myogenic differentiation factor(myoD) of Cyprinus carpio var.Yibu were highly homologous with those of common carp,while myogenin(myoG) was evolutionarily distinctly branched.Using β-actin as the reference gene,quantitative real-time PCR revealed that under still water breeding conditions,the expression levels of myf6 and myoD in Cyprinus carpio var.Yibu were significantly higher than those in the flowing water group(P<0.05),under both breeding modes,the expression of the family member myoG showed no significant difference(P>0.05).There were no significant differences in growth performance,muscle texture,and nutritional characteristics between the two breeding modes(P>0.05).The study indicates that due to the high cost of intensive breeding,the selection of Cyprinus carpio var.Yibu as an object for intensive recirculating aquaculture should be considered carefully.
