摘  要:子宫内膜癌是发达国家和我国部分发达城市女性生殖系统最常见的恶性肿瘤,其发病率呈逐年上升且年轻化趋势。近年来研究表明,免疫逃逸是子宫内膜癌进展的关键机制之一。Janus激酶(Janus Kinase,JAK)/信号转导与转录激活因子(Signal Transducer and Activator of Transcription,STAT)信号通路在调控肿瘤微环境和免疫细胞功能方面发挥重要作用,异常激活的JAK/STAT信号通路通过诱导免疫抑制分子的表达帮助肿瘤细胞规避免疫监视,促进其增殖和转移。同时,胰岛素样生长因子2 mRNA结合蛋白2(Insulin–like Growth Factor 2 mRNA Binding Protein 2,IGF2BP2)作为RNA结合蛋白,能够稳定促癌基因mRNA,进而增强肿瘤细胞的增殖、存活能力,并与JAK/STAT信号通路协同作用,进一步增强肿瘤免疫逃逸能力。中医药在调控肿瘤免疫微环境方面具有多靶点、多途径的优势,能够通过调节JAK/STAT信号通路及相关分子机制抑制肿瘤生长、增强抗肿瘤免疫反应。然而,目前针对JAK/STAT信号通路与IGF2BP2双重靶点的中医药研究仍处于探索阶段,缺乏系统性研究。本研究系统回顾JAK/STAT信号通路与IGF2BP2在子宫内膜癌免疫逃逸中的作用机制,深入探讨中医药调控策略的潜在优势,以期为子宫内膜癌的临床防治提供新的靶点和理论依据。Endometrial cancer is the most common malignant tumor of the female reproductive system in developed countries and some developed cities in China,with an increasing incidence and a trend toward younger patients.Recent studies have shown that immune evasion is a key mechanism in the progression of endometrial cancer.The Janus kinase(JAK)/signal transducer and activator of transcription(STAT)signaling pathway plays a critical role in regulating the tumor microenvironment and immune cell function.Abnormal activation of the JAK/STAT pathway promotes immune evasion by inducing the expression of immunosuppressive molecules,thereby facilitating tumor cell proliferation and metastasis.Additionally,insulin-like growth factor 2 mRNA binding protein 2(IGF2BP2),an RNA-binding protein,stabilizes oncogenic mRNAs,enhancing tumor cell proliferation and survival,and cooperates with the JAK/STAT signaling pathway to further augment immune evasion.Traditional Chinese medicine(TCM)offers multi-target and multi-pathway advantages in regulating the tumor immune microenvironment,and can inhibit tumor growth and enhance antitumor immune responses by modulating the JAK/STAT pathway and related molecular mechanisms.However,research focusing on the dual targeting of the JAK/STAT pathway and IGF2BP2 in TCM is still in its exploratory phase,and systematic studies are lacking.This review aims to provide a comprehensive overview of the role of the JAK/STAT pathway and IGF2BP2 in immune evasion in endometrial cancer and explore the potential regulatory strategies of TCM,in hope of offering new therapeutic targets and theoretical foundations for the clinical prevention and treatment of endometrial cancer.
