摘 要:目的:分析极光激酶A(aurora kinase A,AURKA)在结直肠癌组织中的表达情况及与患者临床病理特征的相关性,预测上游靶基因,探讨其在结直肠癌中的临床意义。方法:生物信息学分析AURKA在结直肠癌组织中的表达情况,免疫组化检测AURKA在结直肠癌及癌旁组织中的表达情况;TargetScan数据库筛选靶向AURKA的miRNA;qPCR检测AURKA与其靶点miRNA在结直肠癌及癌旁组织中的mRNA表达情况,分析两者与临床病理之间的关系。结果:生物信息学分析及免疫组化显示,在结直肠癌中AURKA表达水平明显高于癌旁水平(P<0.05),且与患者脉管侵袭、神经侵袭、Ki-67表达及TP53突变情况相关;通过TargetScan数据库,miR-490-3p可能是AURKA的上游靶基因,两者在结直肠癌中表达呈负相关,miR-490-3p与脉管侵袭、TP53突变相关。结论:AURKA在结直肠癌中高表达,并且可能受miR-490-3p的调控参与结直肠癌的发生发展,有望成为结直肠癌的标志物和治疗靶点。Objective:To analyze the expression of Aurora Kinase A(AURKA) in colorectal cancer tissues and its correlation with clinical pathological features of patients,predict upstream target genes,and explore its clinical significance in colorectal cancer.Methods:Bioinformatics analysis was used to investigate the expression of AURKA in colorectal cancer tissues,and immunohistochemistry was employed to detect the expression of AURKA in colorectal cancer and adjacent tissues.The TargetScan database was screened for miRNAs targeting AURKA.PCR was used to detect the mRNA expression of AURKA and miRNAs in colorectal cancer and adjacent tissues,and analyze the relationship between them and clinical pathology.Results:Bioinformatics analysis and immunohistochemistry revealed that the expression level of AURKA in colorectal cancer was significantly higher than that in adjacent tissues(P<0.05),and was correlated with patient vascular invasion,neural invasion,Ki-67 expression,and TP53 mutation status.Through the TargetScan database,miR-490-3p was identified as a possible upstream target gene of AURKA,and their expressions were negatively correlated in colorectal cancer.miR-490-3p was also associated with vascular invasion and TP53 mutation.Conclusion:AURKA is highly expressed in colorectal cancer and may participate in the occurrence and development of colorectal cancer through the regulation of miR-490-3p,potentially serving as a marker and therapeutic target for colorectal cancer.
