摘 要:肺癌目前是发病率和死亡率第一的癌种,其中非小细胞肺癌(non-small cell lung cancer,NSCLC)占80%~85%。细胞周期失控是恶性肿瘤发生发展的关键因素,包括肺癌在内,约80%的人类肿瘤在细胞周期蛋白D-CDK4/6-INK4-RB通路中表现出异常。CDK4/6抑制剂可通过抑制CDK4/6分子活性阻断肿瘤细胞的细胞周期发挥抗肿瘤效应,当前已在乳腺癌临床治疗中获批使用。从当前CDK4/6抑制剂在非小细胞肺癌中的研究结果看,抗细胞周期是一个较好的新治疗靶点。该文总结了CDK4/6抑制剂抗肿瘤的作用机制,以及当前的一些基础和临床研究的结果,希望为后续以CDK4/6为靶点的抗非小细胞肺癌研究提供新的视野与策略。Lung cancer is currently the most prevalent and fatal type of cancer,with non-small cell lung cancer(NSCLC) accounting for 80% to 85%.Dysregulation of the cell cycle is a critical factor in the initiation and progression of malignancies,including lung cancer,with approximately 80% of human tumors exhibiting abnormalities in the cyclin D-CDK4/6-INK4-RB pathway.CDK4/6 inhibitors exert their antitumor effects by blocking the activity of CDK4/6,thereby interrupting the cell cycle of tumor cells.These agents have already been approved for clinical use in breast cancer treatment.Current research findings regarding CDK4/6 inhibitors in non-small cell lung cancer suggest that targeting the cell cycle represents a promising new therapeutic strategy.This article summarizes the mechanisms by which CDK4/6 inhibitors exert their antitumor effects,as well as some recent preclinical and clinical research outcomes,with the aim of providing new insights and strategies for future investigations targeting CDK4/6 in the treatment of non-small cell lung cancer.
