摘　　要：目的探讨组蛋白去乙酰化酶2(HDAC2)与p21在卵巢上皮性癌(EOC)组织中表达、与临床特征相关性及两者的关系。方法收集2017年3月至2020年3月在漯河市召陵区人民医院95例患者经初次手术治疗切除的癌组织及相应的癌旁组织,分别采用反转录聚合酶链反应(RT-PCR)及蛋白免疫印迹(Western blot)法检测各样本组织中HDAC2、p21的mRNA和蛋白表达水平,分析其表达水平与临床病理特征之间的相关性。结果HDAC2在EOC组织中的表达水平较癌旁组织显著增高(P<0.05),且HDAC2表达量与患者的年龄、肿瘤恶性程度及淋巴转移情况呈正相关;同时,p21在EOC组织中的表达水平较癌旁组织明显降低(P<0.05),且p21表达量与患者的年龄、肿瘤恶性程度及淋巴转移情况呈负相关;EOC组织中HDAC2与p21的表达两者之间呈负相关(相关系数r=-0.707,P<0.001)。结论HDAC2高表达、p21低表达与卵巢上皮性癌的发生、发展、转移、浸润及预后等具有相关性,联合检测HDAC2及p21的表达可能有助于卵巢上皮性癌的临床诊断和治疗。Objective To explore the expression of histone deacetylase 2(HDAC2)and p21 in epithelial ovarian carcinoma(EOC),the clinical features and relationship between them.Methods A total of 95 patients with ovarian epithelial carcinoma treated in the Shaoling District People's Hospital form March 2017 to March 2020 were included.RT-PCR and Western blot methods were used to detect the mRNA and protein expression levels of HDAC2 and p21 in each sample tissue,and the relationship between their expression levels and clinical pathological characteristics,as well as their correlation,was analyzed.Results The expression level of HDAC2 in EOC tissue was significantly higher than that in adjacent tissues(P<0.05)significantly,and it's expression level was positively correlated with the patient's age,tumor malignancy,and lymph node metastasis.Meanwhile,the expression level of p21 in EOC tissue was significantly lower than that in adjacent tissues(P<0.05)significantly,and it's expression level was negatively correlated with the patient's age,tumor malignancy,and lymph node metastasis.There was a negative correlation between the expression of HDAC2 and that of p21 in EOC tissue(r=-0.707,P<0.001).Conclusions High HDAC2 expression and low p21 expression are correlated with the occurrence,development,metastasis,invasion and prognosis of EOC.The combined detection of HDAC2 and p21 expression may be helpful for the clinical diagnosis and treatment of EOC.
