摘  要:拟南芥WUSCHEL RELATED HOMEOBOX 5 (WOX5)在根尖静止中心(QC)特异性表达,其蛋白能从QC细胞移动至小柱干细胞(columella stem cells, CSCs),在根尖干细胞维持中发挥着关键的调控作用。本研究构建了受地塞米松(DEX)诱导控制的WOX5过表达株系,以此建立甲基磺酸乙酯(EMS)诱变的突变体库,并进行了突变体的筛选。结果显示WOX5过表达不仅能诱导根尖产生多层CSC类似细胞,而且还能抑制拟南芥幼苗主根的生长发育。此外,通过筛选获得了46个WOX5过表达表型减弱或不能实现的突变体,其中10个为显性突变体、36个为隐性突变体。通过基因表达分析,我们发现隐性突变体中有些能维持WOX5的过表达状态,而有些WOX5表达量则显著下降。以上研究结果和获得的突变体材料为后续基因定位和WOX5调控机制的研究奠定了良好的基础。WUSCHEL RELATED HOMEOBOX 5(WOX5)is specific expressed in quiescent center(QC)of Ara-bidopsis roots,and its protein can move from QC cells to columella stem cells(cscs).WOx5 plays a key role in the regulation of stem cell maintenance of root tips.In this paper,we constructed the WOx5 overex-pression lines controlled by dexamethasone(DEXx)induction.On the basis,the mutant library of eth-yl-methanesulfonate(EMs)mutagenesis was built to screen the mutants.The results showed that the overexpression of WOx5 could not only induce the generation of multi-layer csc-like cells in root tips,but also inhibit the primary root growth and seedlings development of Arabidopsis.Furthermore,46 mutants were obtained with reduced or unrealized wOx5 overexpression phenotype through screening,of which 10 were dominant mutants and 36 were recessive mutants.Gene expression analysis showed that some of the recessive mutants could maintain the overexpression state of wOx5,while the others had a significant decrease in WOx5 expression.The above studies and the acquisition of mutants laid a good foundation for the subsequent study of gene mapping and wox5 regulatory mechanism.
