摘 要:玉米南方锈病(southern corn rust, SCR)是影响华南地区鲜食玉米生产的主要病害之一,筛选抗南方锈病相关基因对鲜食糯玉米(Zea mays var. sinensis)抗病品种的选育和遗传改良具有重要作用。本研究以鲜食糯玉米高抗南方锈病自交系09N1-1-1和高感病自交系09N9-3为亲本进行杂交,构建F2代分离群体。从F_(2)代群体中挑选高抗和高感南方锈病的单株各30株,分别构建极端性状的DNA混池,通过混合群体分离测序(bulked-segregant analysis sequencing, BSA-Seq)技术初步定位鲜食糯玉米抗南方锈病相关基因。结果表明,测序平均覆盖深度为33.50×,得到1个与抗病相关的候选区域,长度为4.23 Mb,位于玉米10号染色体上,共包含150个基因。其中,有119个基因存在非同义突变, 53个基因存在移码突变。对候选区域内基因进行注释分析,定位到7个与抗病相关的候选基因:Zm00001d023332、Zm00001d023265、Zm00001d023267、Zm00001d023304、Zm00001d023303、Zm00001d023393和Zm00001d023273。研究结果可为鲜食糯玉米南方锈病抗病基因的克隆及抗病新品种选育提供参考依据。Southern corn rust(scR)is one of the major diseases affecting the production of fresh maize in South China region.Screening for ScR resistance genes is of great importance for the breeding and genet-ic improvement of disease-resistant varieties of fresh waxy maize(Zea mays var.sinensis).In this study,the highly SCR-resistant fresh waxy maize inbred line 09N1-1-1 and the highly susceptible inbred line 09N9-3 were used as parents to cross and construct the F_(2) generation segregating populations.From the F2 populations,30 highly ScR-resistant plants and 30 highly susceptible plants were selected to construct two DNA pools with extreme traits and the genes related to SCR resistance in fresh waxy maize were pre-liminarily mapped by the BSA-Seq(bulked-segregant analysis sequencing)technology.The results showed that the average coverage depth of sequencing was 33.50x,and one candidate region related to disease resistance was obtained,which was located on chromosome 10,with a length of 4.23 Mb and a total of 150 genes.Among them,119 genes have non-synonymous mutantions,and 53 genes have frameshift mu-tations.Annotation analysis of the genes in the candidate region mapped 7 candidate genes related to disease resistance:Zm00001d023332,Zm00001d023265,Zm00001d023267,Zm00001d023304,Zm00001d023303,Zm00001d023393 and Zm00001d023273.The results of this study can provide a refer-ence for the cloning of scR resistance genes in fresh waxy maize and the breeding of new disease-resis-tantvarieties.
