摘 要:为了探究干燥方式和电子束辐照(electron beam irradiation,EBI)对天麻多酚的影响,本研究采用真空冷冻干燥、加热减压干燥结合EBI技术处理天麻样品;建立HPLC指纹图谱,同时确定天麻样品中8个多酚物质的含量。通过熵权-逼近理想解排序法(technique for order of preference by similarity to ideal solution,TOPSIS)模型评价天麻品质,确定最优电子束辐照和干燥条件。本研究建立的HPLC法能够准确测定天麻中天麻素、香草酸等8个多酚的含量,分离度均大于1.5,且各组分在相应的浓度范围内与峰面积有良好的线性关系(R 2≥0.9991)。经过熵权-TOPSIS模型分析,天麻通过冷冻干燥并结合20 kGy剂量的电子束辐照后的品质最佳。综上所述,本文建立的HPLC法同时测定8个多酚,方法简便快速,可广泛用于天麻多酚的含量测定,同时确立了天麻最优的电子束辐照和干燥条件,为天麻资源的开发利用提供了理论支持。To investigate the effects of different drying methods and electron beam irradiation(EBI)on the polyphenol content in Gastrodia elata roots,this study employed vacuum freeze-drying and heat-reduced pressure drying in combination with EBI technology.High-performance liquid chromatography fingerprint was established to simultaneously determine the contents of eight polyphenolic compounds,including gastrodin and vanillic acid,in G.elata roots samples.The HPLC method demonstrated excellent resolution for all components,with each pair of adjacent peaks having a resolution greater than 1.5,and showed good linearity(R 2≥0.9991)within the corresponding concentration ranges.The quality of G.elata roots was evaluated using the technique for order of preference by similarity to ideal solution(TOPSIS)model,incorporating entropy weights.The analysis revealed that the optimal conditions for preserving the quality of G.elata roots involved freeze-drying followed by 20 kGy of EBI.In summary,the developed HPLC method provides a simple,rapid,and reliable approach for the simultaneous determination of multiple polyphenols in G.elata roots,and it establishes the optimal EBI and drying conditions,offering theoretical support for the development and utilization of G.elata roots resources.
