摘 要:目的探讨雪莲注射液对心肌缺血再灌注损伤小鼠心功能的影响。方法将30只SPF级雄性C57BL/6小鼠随机分为假手术组、模型组和雪莲注射液组,每组10只。模型组和雪莲注射液组小鼠采用冠状动脉左前降支结扎法建立心肌缺血再灌注损伤模型,假手术组小鼠开胸但不结扎冠状动脉。术后雪莲注射液组给予雪莲注射液4.11 mg/(kg·d)腹腔注射,假手术组和模型组腹腔注射等体积0.9%氯化钠注射液,均1次/d,连续注射7 d。干预结束后,采用超声心动图检测左心室射血分数(LVEF)、左心室短轴缩短率(LVFS)、心排血量(CO)、室间隔增厚率(IVSTP)、左心室舒张末期容积(LVEDV)、左心室收缩末期容积(LVESV),三苯基氯化四氮唑(TTC)法测定心肌梗死面积百分比,末端脱氧核苷酸转移酶介导的缺口末端标记(TUNEL)法测定心肌细胞凋亡指数,微板法测定血清乳酸脱氢酶(LDH)水平,比色法测定血清肌酸激酶(CK)水平,酶联免疫吸附(ELISA)法测定血清白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平。结果与假手术组比较,模型组小鼠LVEF、LVFS、CO、IVSTP均明显降低(P均<0.05),LVEDV、LVESV、心肌梗死面积百分比、心肌细胞凋亡指数及血清CK、LDH、IL-6、TNF-α水平均明显升高(P均<0.05);与模型组比较,雪莲注射液组小鼠LVEF、LVFS、CO、IVSTP均明显升高(P均<0.05),LVEDV、LVESV、心肌梗死面积百分比、心肌细胞凋亡指数及血清CK、LDH、IL-6、TNF-α水平均明显降低(P均<0.05)。结论雪莲注射液对心肌缺血再灌注损伤小鼠心功能具有明显保护作用,这可能与其可抑制炎症反应、细胞凋亡和心室重塑有关。Objective It is to explore the protective effects of Xuelian injection(XLI)on cardiac function of mice with myocardial ischemia-reperfusion injury(MIRI).Methods Thirty SPF male C57BL/6 mice were randomly divided into sham operation group,model group and XLI group,with 10 mice in each group.The mice of the model group and XLI group were modeled by ligating their anterior descending branch of the left coronary artery to establish models of MIRI,and the mice of the sham operation group only underwent thoracotomy without coronary artery ligation.The XLI group was injected intraperitoneally with XLI at a dose of 4.11 mg/(kg·d)after surgery,and the other two groups were injected intraperitoneally with the same volume of 0.9%sodium chloride injection,all once daily,continuously treated for 7 days.After the intervention,the left ventricular ejection fraction(LVEF),left ventricular fractional shortening(LVFS),cardiac output(CO),interventricular septal thickness percent(IVSTP),left ventricular end-diastolic volume(LVEDV)and left ventricular end-systolic volume(LVESV)were measured by echocardiography,the percentage of myocardial infarction(IS/AAR%)was measured by triphenyltetrazolium chloride(TTC),the apoptosis index(AI)of cardiomyocytes was detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL),the serum level of lactate dehydrogenase(LDH)was detected by microplate method,the serum level of creatine kinase(CK)was detected by colorimetric method,and the serum levels of IL-6 and TNF-αwere detected by enzyme linked immunosorbent assay(ELISA).Results Compared with the sham operation group,the LVEF,LVFS,CO,and IVSTP of mice in the model group were significantly lower(all P<0.05),while LVEDV,LVESV,IS/AAR%,AI of cardiomyocytes,and serum levels of CK,LDH,IL-6,and TNF-αwere significantly higher(all P<0.05);compared with the model group,the LVEF,LVFS,CO,and IVSTP of mice in the XLI group were significantly higher(all P<0.05),while LVEDV,LVESV,IS/AAR%,AI of cardiomyocytes,and serum levels of CK,LD
