槲皮素对慢性输卵管炎大鼠上皮间充质转化的影响

摘  要:目的观察槲皮素对慢性输卵管炎大鼠上皮间充质转化的影响,探讨其干预慢性输卵管炎的作用机制。方法将50只SD大鼠随机分为对照组、模型组及槲皮素低、中、高剂量组,每组10只。除对照组外,其余大鼠采用金黄色葡萄球菌悬液复制慢性输卵管炎模型。造模成功后,槲皮素低、中、高剂量组分别给予25 mg/kg、50 mg/kg、100 mg/kg的槲皮素灌胃,对照组和模型组给予等量纯净水灌胃,均1次/d,持续灌胃4周。肉眼观察大鼠输卵管组织形态并进行评分,HE染色观察大鼠输卵管组织病理形态并进行病理评分,Masson染色观察大鼠胶原沉积情况并计算胶原面积占比,实时荧光定量PCR法和Western blot法分别检测输卵管组织中波形蛋白(Vimentin)、Snail mRNA和蛋白表达情况。结果与对照组比较,模型组大鼠出现明显积水、积脓及盆腔粘连样改变,病理可见大量炎性细胞浸润,胶原沉积颜色深,组织形态学评分、病理评分、胶原面积占比和输卵管组织中Vimentin、Snail mRNA及蛋白相对表达量均明显升高(P均<0.05)。与模型组比较,槲皮素各组大鼠输卵管组织病理损伤明显减轻,炎性细胞浸润明显减少,其中槲皮素高、中剂量组大鼠组织形态学评分、病理评分、胶原面积占比和输卵管组织中Vimentin、Snail mRNA及蛋白相对表达量均明显降低(P均<0.05),且以槲皮素高剂量组变化最为明显。结论槲皮素可通过下调慢性输卵管炎大鼠输卵管组织中Vimentin和Snail的表达,抑制上皮间充质转化而发挥治疗作用。Objective It is to observe the effect of quercetin(Que)on epithelial mesenchymal transition in rats with chronic salpingitis,and to investigate its mechanism of action in treating chronic salpingitis.Methods Fifty SD rats were randomly divided into control group,model group,low,medium and high dose groups of Que,with 10 rats in each group.The rats of all groups except for the control group were modeled to establish models of chronic salpingitis by staphylococcus aureus suspension.After successful modeling,the low,medium and high dose groups of Que were given 25 mg/kg,50 mg/kg and 100 mg/kg of Que by gavage,respectively,and the control and model groups were given equal amounts of pure water by gavage,all once daily,continuously treated for 4 weeks.The morphology of oviduct tissue of the rats was observed by naked eye and was scored,the pathological morphology of oviduct tissue of the rats was observed by HE staining and were scored,the collagen deposition was observed by Masson staining and the percentage of collagen area was calculated,and the mRNA and protein expressions of vimentin,Snail in oviduct tissue were respectively detected by real-time fluorescence quantitative PCR and Western blot.Results Compared with the control group,rats in the model group showed obvious hydrosalpinx,pyosalpinx and pelvic adhesion-like changes,with pathologic changes such as a large number of inflammatory cell infiltration,deep color of collagen deposition,and the histomorphometric scores,pathological scores,collagen area percentage and relative mRNA and protein expressions of vimentin,Snail in oviductal tissues were all significantly increased(all P<0.05).Compared with the model group,the pathological damage of fallopian tube tissues of rats in all Que groups was significantly improved,and the inflammatory cell infiltration was significantly reduced,among which the histomorphological score,pathological score,collagen area percentage,and relative mRNA and protein expressions of vimentin,Snail in fallopian tube tissues of rats in t
