摘 要:【目的】旨在研究巨魾种质资源、物种间的亲缘关系及遗传多样性。【方法】以云南省3条水系(元江、怒江、澜沧江)5个种群共60尾巨魾为研究对象,选取线粒体ND6基因作为分子标记,对巨魾遗传多样性进行研究。【结果】巨魾线粒体ND6基因序列全长525 bp,60条序列获得41种单倍型;41种单倍型序列中A、T、C、G平均含量分别为40.0%、13.7%、34.8%、11.5%,A+T含量为53.7%,C+G的含量为46.3%;共检测到143个变异位点,375个保守位点,114个简约信息位点;核苷酸多样性和单倍型多样性分别为0.08769±0.00235和0.960±0.017;41种单倍型间平均遗传距离为0.101。【结论】巨魾41种单倍型与其他7种鱼类ND6基因序列用NJ法构建系统发育树,发现巨魾单独聚为一支。【Objective】This study aims to investigate the germplasm resources,genetic relationship,and genetic diversity of Bagarius yarrelli.【Method】The genetic diversity of B.yarrelli was studied by using mitochondrial ND6 gene of 60 individuals as a molecular marker in five populations of Bagariusyarrelli from the three rivers(Yuanjiang,Nujiang,and Lancangjiang)in Yunnan Province.【Result】The results showed that the total length of mitochondrial ND6 was 525 bp,and 41 haplotypes were defined from 60 sequences.The average contents of A,T,C and G in all samples was 40.0%,13.7%,34.8%,and 11.5%,respectively.The A+T content was 53.7%and the percentage of C+G content was 46.3%.A total of 143 mutation sites,375 conserved sites,and 114 parsimony informative sites were detected.The nucleotide diversity and haplotype diversity were 0.08769±0.00235 and 0.960±0.017,respectively;and the average genetic distance between 41 haplotypes in 60 individuals was 0.101.【Conclusion】A phylogenetic tree was constructed using the Neighbor-Joining(NJ)method for the ND6 gene sequence of 41 haplotypes of B.yarrelli and 7 other species of fish.It was found that B.yarrelli clustered into a single branch.
