节节麦花粉离体萌发体系的建立

摘  要:【目的】节节麦作为普通小麦的重要育种资源,其利用方式主要是通过杂交技术,杂交成功的关键在于花粉活力。花粉体外萌发技术是花粉活力测定中常用的技术之一。通过构建节节麦花粉离体萌发体系,可以为节节麦在小麦育种中的应用提供了有力的技术支持。【方法】以节节麦(ssp.tauschii亚种)为材料,12种花粉离体萌发培养体系为候选方法,研究了不同浓度组合的棉子糖、硼酸、氯化钙和pH对节节麦花粉离体萌发的影响,从而找到适合的离体萌发培养体系。【结果】节节麦花粉在594 mmol/L棉子糖+0.81 mmol/L硼酸+2.04 mmol/L氯化钙+3%明胶,pH=5.8时,于28℃条件下培养30 min,可诱导萌发率高达75%以上。【结论】试验培养基适用于节节麦花粉离体萌发,能够准确检测节节麦花粉活性。【Objective】Aegilops tauschii Cosson is an important breeding resource for common wheat,and its utilization primarily relies on hybridization techniques.The key to successful hybridization is pollen viability.In vitro pollen germination is one of the commonly used techniques for determining pollen viability.This study aimed to establish an effective in vitro germination system to promote the utilization of A.tauschii Cosson in wheat breeding.【Method】The effects of different concentration combinations of raffinose,boric acid,calcium chloride and pH on the in vitro germination of pollen were investigated using A.tauschii Cosson(ssp.tauschii subspecies)was used as the experimental material,and twelve pollen in vitro germination culture systems were evaluated as candidate methods to identify a suitable in vitro germination culture system.【Result】The results showed that when pollen was cultured at 28℃for 30 minutes at a pH of 5.8 in a medium containing 594 mmol/L raffinose,0.81 mmol/L boric acid,2.04 mmol/L calcium chloride,and 3%gelatin,the germination rate could be induced to reach 75%or higher.【Conclusion】The above medium was found to be suitable for the in vitro germination of pollen and can accurately detect the pollen activity of A.tauschii Cosson.
