摘 要:本研究旨在建立改良3,5-二硝基水杨酸(DNS)还原性糖测量方法。以无水葡萄糖为标准品,采用常温下3,5-二硝基水杨酸(DNS)显色试剂结合一定浓度的高锰酸钾(KMnO4),2000 r·min^(-1)涡旋混匀15 min建立的改良DNS法进行还原糖显色,并进行相应方法学研究。结果表明,使用上述的改良DNS还原性糖测量方法,测量波长为540 nm,以无水葡萄糖为标准品的线性回归方程为Y=15.16X+0.0244(R^(2)=0.9899),在0.00703~0.05621 g·mL^(-1)范围内线性关系良好;相同浓度标准品平行测试完成的精密度实验显示RSD为3.29%;以苹果样品为测试对象的加样回收率为(100.86±2.37)%。苹果样品还原糖浓度测试、加样回收率等结果与NY/T 2742标准中DNS法测定结果无明显差异(P>0.05)。改良DNS还原糖分光光度测量法简便快捷,灵敏度、精密度和稳定性良好,可以作为还原糖含量测定的方法,具有良好的应用前景。The aim of this study was to establish modified 3,5-dinitrosalicylic acid(DNS)method for measuring reducing sugar concentration.Improved DNS method was established by 3,5-dinitrosalicylic acid(DNS)as basic chromogenic reagent,,combined with certain concentration of potassium permanganate(KMnO4),vortexing and mixing at 2000 r·min^(-1) for 15 minutes.Methodological studies on improved DNS spectrophotometry measure of reducing sugar were conducted with anhydrous glucose as standard.The results show that wavelength of Improved DNS Measurement on reducing sugar was 540 nm.The linear regression equation of anhydrous glucose as standard was Y=15.16X+0.0244(R^(2)=0.9899),and the linear relationship was good in the range of 0.00703~0.05621 g·mL^(-1).RSD of precision experiments performed in parallel tests of standards with same concentration was 3.29%.The recoveries of apple samples were(100.86±2.37)%.There was no significant difference between the results of reducing sugar concentration test and sample recovery rate of apple samples and the results of DNS method in NY/T 2742 standard(P>0.05).Method of Improved DNS Spectrophotometry Measurement on reducing sugar were simple and fast,with good sensitivity,precision and stability,and can be used as a method for the detection of reducing sugar content,which has a good application prospect.
