摘  要:目的研究miR-10b、HOXA1及PI3K/AKT在低级别子宫颈鳞状上皮内病变(LSIL)、高级别子宫颈鳞状上皮内病变(HSIL)和早期宫颈癌患者宫颈组织中的表达水平的差异,探讨宫颈癌发病机制。方法选取2021年7月至2021年12月福建省第二人民医院门诊就诊,经宫颈防癌筛查三阶梯程序,宫颈病理检查确诊为LSIL、HSIL和早期宫颈癌的新发患者各10例。采用qPCR方法检测miR-10b表达水平,采用蛋白质印迹法检测HOXA1表达水平,采用免疫组织化学染色法检测PI3K/AKT表达水平,分析三者之间可能存在的关系。结果miR-10b表达量在LSIL、HSIL和早期宫颈癌患者中呈逐渐下降(P<0.01),HOXA1蛋白表达量在LSIL、HSIL和早期宫颈癌患者中呈逐渐上升(P<0.01),PI3K/AKT阳性率在LSIL、HSIL和早期宫颈癌患者中亦呈逐渐上升趋势,但组间差异无统计学意义(P>0.05)。miR-10b表达量与HOXA1蛋白表达量、PI3K/AKT表达水平均呈负相关关系(P<0.01),HOXA1蛋白表达量与PI3K/AKT表达水平均呈正相关关系(P<0.01)。结论在宫颈癌病变过程中,miR-10b可能是通过调控HOXA1/PI3K/AKT通路发挥抑癌基因功能。Objective To investigate the differences in the expression levels of miR-10b,Hoxa1 and PI3K/AKT between cervical tissues from LSIL,HSIL and early-stage cervical cancer patients.Methods A total of 10 newly diagnosed patients with LSIL,HSIL and early cervical cancer were selected from the outpatient department of Fujian Second People’s Hospital from July 2021 to December 2021 through the three-step cervical cancer screening procedure.qPCR was used to detect the expression level of miR-10b,western blot was used to detect the expression level of HOXA1,and immunohistochemical staining was used to detect the expression level of PI3K/AKT,and the possible relationship between the three was analyzed.Results The expression level of miR-10b was gradually decreased in patients with LSIL,HSIL and early cervical cancer(P<0.01),and the expression level of HOXA1 protein was gradually increased in patients with LSIL,HSIL and early cervical cancer(P<0.01).The positive rate of PI3K/AKT also increased gradually in patients with LSIL,HSIL and early cervical cancer,but there was no statistical significance between groups(P>0.05).The expression level of miR-10b was negatively correlated with HOXA1 protein expression level and PI3K/AKT expression level(P<0.01),and the expression level of HOXA1 protein was positively correlated with PI3K/AKT expression level(P<0.01).Conclusions In cervical cancer lesions,miR-10b may function as a tumor suppressor gene by regulating the Hoxa1/PI3K/AKT pathway.
