榄香烯对不同类型乳腺癌细胞系的影响机制研究概况

摘  要:乳腺癌是女性最常见的恶性肿瘤,严重威胁女性的健康,榄香烯是从温郁金、温莪术、姜黄等中药提取分离得到的倍半萜烯类化合物,其可以诱导乳腺癌肿瘤细胞的凋亡,阻滞细胞周期的进展,影响肿瘤微环境,以及细胞糖代谢等。对于不同类型的乳腺癌细胞系,榄香烯作用机制亦不同。Her-2阳性乳腺癌,榄香烯和曲妥珠单抗联合使用可以提高患者的治疗效果,提高患者的免疫力,减轻心脏毒性。榄香烯还可以逆转多种乳腺癌细胞的耐药,与多种化疗药物联合应用还可以起到协同增敏的作用。对于三阴性乳腺癌细胞系,榄香烯可以通过抑制肿瘤细胞的糖酵解,从而改变肿瘤微环境,达到抑制肿瘤生长的作用;还可以通过胰岛素样生长因子1(IGF1/IGF1R)途径,诱导细胞凋亡,榄香烯还可以诱导肿瘤细胞发生自噬;对于雌激素受体阳性乳腺癌细胞系MCF-7,榄香烯除了可以抑制肿瘤细胞的糖酵解,还可以抑制雷帕霉素靶蛋白(mTOR)信号通路引发细胞发生自噬引起细胞凋亡。榄香烯还可以通过ER/MTA3/Snail/E-cadherin通路,抑制细胞的迁移和侵袭,阻滞细胞周期进展。本文就榄香烯对不同类型乳腺癌细胞系的影响及机制进行综述。Breast cancer is the most common malignant tumor in women,which seriously threatens women's health.Elemene is a sesquiterpene compound extracted from Curcuma wenyujin,Curcuma wenzedoaria,turmeric and other traditional Chinese medicines.It can induce apoptosis of breast cancer tumor cells,block the progress of cell cycle,and affect tumor microenvironment and cell glucose metabolism.The mechanism of elemene is different for different types of breast cancer cell lines.For Her-2 positive breast cancer,the combination of elemene and trastuzumab can improve the therapeutic effect of patients,improve their immunity,and reduce cardiac toxicity.Elemene can also reverse the drug resistance of multiple breast cancer cells,and can also play a synergistic role in sensitizing when used with multiple chemotherapy drugs.For triple negative breast cancer cell lines,elemene can change the tumor microenvironment by inhibiting the glycolysis of tumor cells,thus achieving the effect of inhibiting tumor growth.Apoptosis can also be induced through the insulin-like growth factor 1(IGF1/IGF1R)pathway,and elemene can also induce autophagy in tumor cells.For estrogen receptor positive breast cancer cell line MCF-7,elemene can not only inhibit the glycolysis of tumor cells,but also inhibit the autophagy of cells caused by the rapamycin target protein(mTOR)signal pathway,leading to apoptosis.Elemene can also inhibit cell migration,invasion,and cell cycle arrest through the ER/MTA3/Snail/E-cadherin pathway.This article reviewed the effects and mechanisms of elemene on different types of breast cancer cell lines.
