摘  要:目的 探讨单细胞测序联合批量转录组测序方法对肺腺癌(LUAD)中CD8+T细胞相关标志物鉴定效果。方法 通过加权基因共表达网络分析(WGCNA)筛选与生存时间密切相关的预后细胞相关标志物。使用富集分析注释预后细胞相关标志物的生物学机制。通过蛋白质相互作用(PPI)网络,耐药分析和免疫治疗反应分析鉴定出预后细胞相关标志物中的Hub基因。结果 利用GSE131907和GSE149655单细胞数据集,系统地识别出LUAD中的CD8+T细胞亚群,并筛选出共同高表达的CD8+T细胞相关标志物(CD8TRBs)。WGCNA分析表明这些CD8TRBs与LUAD患者的临床特征(包括生存状态、T分期和N分期)显著相关。富集分析揭示CD8TRBs参与的多种免疫相关通路。通过PPI网络分析和MCC算法,确定了CD8TRBs的Hub基因,并证实这些基因的表达水平与LUAD患者的生存时间和治疗效果密切相关。结论 本研究鉴定了CD8+T细胞相关的生物标志物,为预测患者的预后和优化个体化治疗策略提供了新的可能性。Objective To investigate the effect of single cell sequencing combined with batch transcriptome sequencing on the identification of CD8+T cell-related markers in lung adenocarcinoma(LUAD).Methods This study combined single cell sequencing and batch transcriptome sequencing to identify CD8+T cell-associated markers in lung adenocarcinoma.Weighted gene co-expression network analysis(WGCNA)was used to screen prognostic markers closely related to survival time.Annotation of biological mechanisms of prognostic cell-related markers using enrichment analysis.Hub genes in prognostic cell-related markers were identified by protein interaction(PPI)networks,drug resistance analysis,and immunotherapy response analysis.Results CD8+T cell subsets in LUAD were systematically identified using the GSE131907 and GSE149655 single-cell datasets,and CD8+T cell-associated markers(CD8TRBs)that were jointly highly expressed were screened.WGCNA analysis showed that these CD8TRBs were significantly associated with clinical characteristics of LUAD patients,including survival status,T stage,and N stage.Enrichment analysis revealed that CD8TRBs were involved in a variety of immune-related pathways.Through PPI network analysis and the MCC algorithm,the hub genes of CD8TRBs were identified,and it was confirmed that the expression levels of these genes were closely related to the survival time and treatment effect of LUAD patients.Conclusions This study identifies biomarkers associated with CD8+T cells,providing new possibilities for predicting patient prognosis and optimizing individualized treatment strategies.
