摘  要:为优化糯青稞中β-葡聚糖提取率,基于单因素试验分析结果,结合响应面(Box-behnken)试验设计原则,以pH值、料液比、提取时间和提取温度4个因素进行响应面分析,并据此建立了β-葡聚糖提取率的二次回归方程。以β-葡聚糖提取率为指标,通过响应面法确定了最佳提取工艺条件。结果表明,最佳提取工艺参数为pH值6.94,料液比1∶13.94(g∶mL),提取时间2.47 h,提取温度38.82℃。β-葡聚糖提取率的理论值为3.63%,而根据实际情况,最佳提取条件是料液比1∶15(g∶mL),提取温度40℃,提取时间2.5 h,在此条件下提取率可达到3.72%,为糯青稞中β-葡聚糖的提取工艺研究提供了科学依据。The purpose of this study was to optimize the extraction technology ofβ-glucan from waxy hull-less barley.Based on the results of single factors,the optimum extraction conditions ofβ-glucan were obtained by the Box-behnken response surface.Four factors and three levels of pH,solid-liquid ratio,extraction time and temperature were selected.Quadratic regression equation ofβ-glucan extraction was established,and the extraction rate was chose as the index in this study,and obtained the optimum extraction conditions.The results showed that a theoretical maximum polysaccharide yield of 3.63%was achieved at pH 6.94,a solid-liquid ratio of 1∶13.94(g∶mL)for 2.47 h at 38.82℃.According to the actural situation,the optimal process conditions were solid-liquid ratio of 1∶15(g∶mL),extraction temperature of 40℃,extraction time of 2.5 h,and theβ-glucan yield was 3.72%under this condition.The results of this study provided scientific reference for the extraction ofβ-glucan from waxy hull-less barley.
