摘　　要：目的分析氧连氮-乙酰葡萄糖胺(O-GlcNAc)转移酶(OGT)在肝细胞癌中的表达水平及其临床意义。方法选取2022年7月-2023年12月石河子大学第一附属医院肝胆外科20例肝细胞癌患者、20例肝硬化患者及20名健康体检者。采用蛋白质印迹法检测肝细胞癌与瘤旁组织中OGT表达水平,采用酶联免疫吸附法检测血清OGT表达水平,比较肝细胞癌患者肝细胞癌组织与癌旁组织OGT蛋白表达水平及肝细胞癌患者、肝硬化患者、健康体检者血清OGT水平。结果肝细胞癌组织OGT蛋白表达水平高于癌旁组织(P<0.05);肝细胞癌患者与肝硬化患者血清OGT水平均高于健康体检者(P<0.05);肝细胞癌及肝硬化患者血清OGT水平比较,差异无统计学意义(P>0.05)。结论肝细胞癌组织中OGT表达水平较高,检测OGT表达水平可指导临床诊断肝细胞癌,有望作为肝细胞癌的重要诊断靶点。Objective To analyze the expression level and clinical significance of O-linked-β-N-acetylglucosamine(O-GlcNAc)transferase(OGT)in hepatocellular carcinoma.Methods A total of 20 patients with hepatocellular carcinoma,20 patients with liver cirrhosis,and 20 healthy subjects were selected from the Department of Hepatobiliary Surgery,The First Affiliated Hospital of Shihezi University from July 2022 to December 2023.The expression level of OGT in hepatocellular carcinoma and adjacent tissues was detected by Western blotting,and the expression level of serum OGT was detected by enzyme-linked immunosorbent assay.The expression levels of OGT protein in hepatocellular carcinoma tissues and the adjacent tissues of hepatocellular carcinoma patients were contrasted.Additionally,the serum OGT levels among hepatocellular carcinoma patients,liver cirrhosis patients,and healthy individuals were compared.Results The expression level of OGT protein in hepatocellular carcinoma tissue was higher than that in adjacent tissues(P<0.05).The serum OGT levels of liver cancer patients and cirrhosis patients were higher than those of the healthy physical examination group(P<0.05).There was no statistically significant difference in serum OGT levels between patients with liver cancer and cirrhosis(P>0.05).Conclusions The expression level of OGT is relatively high in hepatocellular carcinoma tissue,and detecting the expression level of OGT can guide clinical diagnosis of hepatocellular carcinoma,which is expected to be an important diagnostic target for hepatocellular carcinoma.
