摘 要:目的:抗原处理相关转运蛋白1(transporter associated with antigen processing 1,TAP1)与自身免疫性疾病、自身炎症性疾病和癌症等多种疾病关系密切,但其与胃腺癌的关系尚不明确。本研究旨在探讨胃腺癌中TAP1的表达及其意义。方法:通过基因型-组织表达(genotype-tissue expression,GTEx)、癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据库及免疫组织化学方法检测TAP1在胃腺癌中的表达并分析其与病理参数的相关性,使用R软件pROC包进行受试者操作特征(receiver operating characteristic,ROC)曲线分析;使用Kaplan-Meier Plotter在线分析网站探讨TAP1与胃腺癌预后的相关性;使用肿瘤免疫估算工具(tumor immune estimation resourse,TIMER)分析TAP1与免疫细胞浸润之间的相关性;在基因表达谱互动分析(Gene Expression Profiling Interactive Analysis,GEPIA)数据库中筛选胃腺癌前100个与TAP1共表达相关的基因,进行基因本体(Gene Ontology,GO)和京都基因和基因组数据库(Kyoto Encyclopedia of Genes and Genomes,KEGG)富集分析。结果:与正常组织相比,胃腺癌组织中TAP1蛋白和mRNA表达水平均显著上调(均P<0.05)。诊断ROC曲线显示曲线下面积(area under the curve,AUC)为0.826。TAP1的表达水平与胃腺癌患者的肿瘤分级有关(P<0.01),与病理分期、年龄均无关(均P>0.05)。Kaplan-Meier生存曲线显示TAP1高表达患者总生存期、无进展生存期较低表达患者均延长(均P<0.05)。富集分析显示TAP1表达与T细胞受体结合、主要组织相容性复合物(major histocompatibility complex,MHC)蛋白生成、抗原加工与提呈等过程密切相关。免疫微环境中CD4~+T淋巴细胞等细胞浸润及免疫检查点基因的表达与TAP1的表达水平均呈正相关(均P<0.05)。结论:TAP1在胃腺癌中高表达,具有较好的诊断效能,与较好的胃腺癌预后相关。TAP1可能参与T细胞受体结合、抗原加工与提呈等生物学过程,并且与多种免疫细胞浸润高�Objective:The transporter associated with antigen processing 1(TAP1)is closely related to autoinflammatory diseases,autoinflammatory disease,and cancers,but its relationship with gastric adenocarcinoma remains unclear.This study aims to explore the expression of TAP1 in gastric adenocarcinoma and its significance.Methods:The expression of TAP1 in gastric adenocarcinoma was detected using the Genotype-Tissue Expression(GTEx)and The Cancer Genome Atlas(TCGA)databases and immunohistochemistry.The correlation between TAP1 expression and pathological parameters was analyzed,and receiver operating characteristic(ROC)curve analysis was performed using the R software pROC package.The correlation between TAP1 expression and the prognosis of GAC was assessed through the Kaplan-Meier Plotter online analysis tool.The correlation between TAP1 expression and immune cell infiltration was analyzed using the Tumor Immune Estimation Resource(TIMER).Additionally,the top 100 genes co-expressed with TAP1 in gastric adenocarcinoma were identified through the Gene Expression Profiling Interactive Analysis(GEPIA)database,followed by Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis.Results:Compared to normal tissue,TAP1 protein and mRNA expression levels were significantly upregulated in gastric adenocarcinoma tissues(both P<0.05).The diagnostic ROC curve showed an area under the curve(AUC)of 0.826.TAP1 expression levels were correlated with tumor grading(P<0.01)but were not related to pathological staging or age(both P>0.05).Kaplan-Meier survival curves showed that patients with high TAP1 expression had significantly longer overall survival than those with low TAP1 expression(both P<0.05).Enrichment analysis revealed that TAP1 expression is closely related to T cell receptor binding,major histocompatibility complex(MHC)protein complex formation,and antigen processing and presentation.The expression of immune cell infiltration,such as CD4+T lymphocytes,was positively correlated with TAP1 expression(al
