摘 要:为了克服体外癌细胞系或细胞系衍生的异体移植(cell-derived tumour xenograft,CDX)模型的局限性,并产出更可靠的临床前结果,科研人员引入了患者来源癌症模型(patient-derived models of cancer,PDMC)。目前,PDMC主要包括两大类,即患者来源异种移植(patient-derived xenograft,PDX)模型和患者来源类器官(patient-derived organoids,PDO)模型。PDX和PDO模型各具特色与优势,由于PDMC与患者肿瘤特征的高度一致性,已被广泛采纳于各类肿瘤研究,尤其在抗肿瘤药物的研发和筛选、个性化医疗和精准医疗的开发领域取得了显著的研究进展。近年来,中国甲状腺癌发病率持续攀升,已成为亟待解决的公共卫生的挑战。因此,构建全面和有效的临床前甲状腺癌模型,以探索新型治疗药物和治疗方法,显得尤为迫切与重要。随着技术的不断进步和模型的持续优化,PDMC有望为甲状腺癌及其他肿瘤疾病的精准治疗提供更加坚实的实验基础和个体化治疗策略。To overcome the limitations of in vitro cancer cell lines or cell derived xenograft(CDX)models,and to produce more reliable preclinical results,researchers have introduced patient-derived models of cancer(PDMC).Currently,PDMC mainly include two types:patient-derived xenograft(PDX)model and patient-derived organoids(PDO)model.Both PDX and PDO models have distinct characteristics and advantages.Due to the high consistency between PDMC and the characteristics of patient’s tumors,these models have been widely adopted in various cancer research areas,particularly in the development and screening of anti-cancer drugs,as well as in personalized medicine and precision medicine,showing significant research progress.In recent years,the incidence of thyroid cancer in China has been steadily increasing,becoming an urgent public health challenge.Therefore,establishing comprehensive and effective preclinical thyroid cancer models to explore novel therapeutic drugs and treatment methods is especially urgent and important.With continuous technological advancements and optimization of models,PDMC are expected to provide a solid experimental foundation and individualized treatment strategies for precision therapy in thyroid cancer and other tumors.
