HE染色切片部分区域褪色后行免疫组织化学染色的方法探讨

摘　　要：目的在某些微小标本中,进行褪色再染色时,保留部分原始HE染色非常关键。本研究探讨了使用UV光固化胶对标本进行区域性保护,以保留原始HE染色后,再进行免疫组化染色的应用,以评估其在病理科染色中的效果。方法:切片去除盖玻片后,对需要保留的部分使用UV光固化胶进行封闭保护,再对剩余部分进行褪色,并随后进行免疫组化染色。观察并比较保护区域与染色区域的效果。结果经过UV光固化胶保护后,原始HE染色得以完整保留,且免疫组化染色效果良好。结论UV光固化胶能够有效保护原始HE染色,并且不会影响后续染色操作。该方法为一种新型染色技术,值得在病理科染色中推广应用。Objective In certain small specimens,it is crucial to retain part of the original HE staining during re-staining after discoloration.This study explores the use of UV-curable resin for regional protection of specimens to preserve the original HE staining before performing immunohistochemical staining,aiming to assess its effectiveness in pathological staining procedures.Methods After removing the coverslip from the slides,UV-curable resin was applied to protect the areas that needed to be preserved,followed by discoloration of the remaining regions and subsequent immunohistochemical staining.The effects of the protected and stained regions were observed and compared.Results After protection with UV-curable resin,the original HE staining was well preserved,and the immunohistochemical staining yielded good results.Conclusion UV-curable resin effectively protects the original HE staining without interfering with subsequent staining procedures.This method is a novel staining technique with promising potential for use in pathological laboratories.
