黄芪配伍熟地黄对失神经大鼠肌萎缩诱导骨质疏松症影响

摘  要:目的观察黄芪配伍熟地黄对失神经大鼠肌萎缩诱导的骨质疏松症的影响。方法采用切断右侧坐骨神经结合肌肉注射地塞米松的方法建立失神经肌萎缩合并骨质疏松大鼠模型。将造模后的48只SD大鼠随机分为假手术组、模型组、阳性药组、黄芪组、熟地黄组、黄芪-熟地黄组,分别给予相应的药物灌胃,连续给药2个月后处死动物。测定大鼠两侧腓肠肌湿重比;HE染色观察腓肠肌和股骨远端病理变化;micro CT检测大鼠股骨远端骨微结构;根据试剂盒说明测定大鼠血清大鼠肌肉生长抑制素(MSTN)、胰岛素样生长因子1(IGF-1)、碱性磷酸酶(AKP)、Ⅰ型胶原羟基端肽β降解产物(β-CTX)、超氧化物歧化酶(SOD)、丙二醛(MDA)含量;免疫组化染色检测大鼠腓肠肌肌肉特异性环指蛋白1(MuRF1)和生肌决定因子1(MyoD1)的阳性表达水平。结果与模型组相比,黄芪-熟地黄组能明显提高大鼠腓肠肌湿重比(P<0.01),降低大鼠血清MSTN、AKP、β-CTX、MDA含量,同时提高IGF-1、SOD含量,除SOD外差异均具有统计学意义(P<0.05,P<0.01)。腓肠肌HE染色结果显示,与模型组比较,黄芪-熟地黄组大鼠腓肠肌肌纤维明显排列规则、紧密,肌间隙减小,肌纤维增粗,肌细胞核增多、增大;股骨HE染色和mirco CT结果均显示黄芪-熟地黄组大鼠股骨骨小梁数目明显增多,厚度增加,骨小梁排列较紧密,间隙减小。免疫组化染色结果显示,与模型组比较,黄芪-熟地黄组大鼠腓肠肌MuRF1的表达水平明显降低(P<0.01),MyoD1的表达水平明显增强(P<0.01)。结论黄芪配伍熟地黄能有效改善失神经大鼠肌萎缩诱导的骨质疏松症,这可能与黄芪配伍熟地黄降低骨骼肌氧化应激水平,抑制泛素-蛋白酶体途径,并促进肌卫星细胞增殖分化有关。Objective To observe the effect of Huangqi(Astragali Radix)combined with Shudihuang(Rehmanniae Radix Praeparata)on muscle atrophy induced osteoporosis in denervated rats.Methods A rat model of denervated muscle atrophy combined with osteoporosis was established by cutting the right sciatic nerve and injecting dexamethasone into the muscle.And then 48 SD rats were randomly divided into the sham surgery group,the model group,the positive drug group,the Huangqi group,the Shudihuang group,and the Huangqi-Shudihuang group.The corresponding drugs were given by gavage,and the rats were killed after continuous administration for 2 months.The wet-to-weight ratio of gastrocnemius muscle tissues was measured.Pathological changes of gastrocnemius and distal femur were observed by HE staining.The microstructure of distal femur bone in rats was detected by using micro CT.The contents of serum MSTN,IGF-1,AKP,β-CTX,SOD and MDA were detected according to the reagent kit instructions.The positive expression levels of MuRF1 and MyoD1 in the rat gastrocnemius muscle were detected by immunohistochemical staining.Results Compared with the model group,Huangqi-Shudihuang could significantly increase the wet-to-weight ratio of gastrocnemius muscle of rats(P<0.01),decrease the contents of MSTN,AKP,β-CTX and MDA in serum,and increase the contents of IGF-1 and SOD in serum,with statistical significance except SOD(P<0.05,P<0.01).HE staining of gastrocnemius muscle showed that,compared with the model group,the muscle fibers of gastrocnemius muscle in the Huangqi-Shudihuang group were arranged regularly and tightly,the muscle space was reduced,the muscle fibers were thickened,and the muscle nucleus was increased.Both HE staining of femur and mirco CT showed that the number and thickness of bone trabeculae of femur in the Huangqi-Shudihuang group were significantly increased,the trabeculae were closely arranged,and the space was reduced.Immunohistochemical staining showed that,compared with the model group,Huangqi-Shudihuang group could signi
