摘 要:目的:借助网络药理学及分子对接技术来探究防己黄芪汤在治疗糖尿病肾病(diabetic kidney disease,DKD)方面的潜在的作用机制。方法:我们借助TCMSP和ETCM两种数据库筛选防己黄芪汤的有效化学组分和与其相关联的作用靶点;然后利用人类基因数据库(GeneCards)搜索DKD疾病的相关基因靶点。将疾病靶点与药物靶点取交集并利用String平台构建交集靶点的蛋白质—蛋白质间相互作用(PPI)网络。将PPI网络的TSV文件导入Cytoscape3.7.1软件中,根据度值来设定靶点的形状大小和明暗程度以直观展示核心靶点;利用Metascape数据库对成药-疾病交集靶点进行GO和KEGG富集分析,根据分析结果得到防己黄芪汤治疗DKD的主要生物进程与通路。最后使用Autodock软件和Pymol软件进行分子对接模拟和可视化展示。结果:得到防己黄芪汤的潜在作用靶点235个,DKD的潜在靶点3665个,两者的交集靶点179个,经筛选得到的核心靶点有肿瘤蛋白p53(tumor protein p53,TP53)、丝氨酸/苏氨酸蛋白激酶(AKT serine/threonine kinase 1,AKT1)等;GO功能富集分析与KEGG通路富集分析结果包括:对激素的反应、对无机物的反应、脂质和动脉粥样硬化、糖尿病并发症中的AGE-RAGE信号通路等。分子对接结果显示TP53与槲皮素及山奈酚、AKT1与槲皮素及山奈酚结合活性均良好。结论:防己黄芪汤可以通过干预多基因、多靶点、多通路来治疗DKD。它可以改善血糖血脂的代谢,减轻胰岛素抵抗,缓解炎症反应及氧化应激,改善肾脏血流动力学等,从多种方面起到治疗DKD的作用。Objective:To explore the potential mechanism of Fangji Huangqi Decoction in the treatment of diabetic kidney disease(DKD)by using network pharmacology and molecular docking technology.Methods:We screened the effective chemical components of Fangji Huangqi Decoction and their associated targets using TCMSP and ETCM databases;then used the human gene database(GeneCards)to search for DKD disease-related gene targets.The disease targets and drug targets were intersected and the protein-protein interaction(PPI)network of the intersection targets was constructed using the String platform.The TSV file of the PPI network was imported into Cytoscape3.7.1 software,and the shape and brightness of the targets were set according to the degree value to intuitively display the core targets;the GO and KEGG enrichment analysis of the drug-disease intersection targets was performed using the Metascape database,and the main biological processes and pathways of Fangji Huangqi Decoction in the treatment of DKD were obtained based on the analysis results.Finally,the molecular docking simulation and visualization were performed using Autodock software and Pymol software.Results:235 potential targets of Fangji Huangqi Decoction and 3665 potential targets of DKD were obtained,and 179 targets overlapped with the two.The core targets obtained by screening included tumor protein p53(TP53),serine/threonine kinase 1(AKT serine/threonine kinase 1,AKT1),etc.The results of GO functional enrichment analysis and KEGG pathway enrichment analysis included:response to hormones,response to inorganic substances,lipids and atherosclerosis,AGE-RAGE signaling pathway in diabetic complications,etc.Molecular docking results showed that TP53 had good binding activity with quercetin and kaempferol,and AKT1 had good binding activity with quercetin and kaempferol.Conclusion:Fangji Huangqi Decoction can treat DKD by intervening in multiple genes,multiple targets,and multiple pathways.It can improve blood sugar and blood lipid metabolism,reduce insulin resistance
