真武汤治疗眩晕的潜在机制探究

摘  要:目的:通过网络药理学和分子对接技术探讨真武汤治疗眩晕的潜在机制。方法:将真武汤组成药物分别通过TCMSP和UniProt数据库获取活性成分和潜在靶点;利用GeneCards、DRUGBANK和OMIM数据库获取眩晕疾病的相关靶点;绘制韦恩图并得到交集靶点,通过STRING数据库,生成PPI网络,筛选关键靶点位并实现可视化;通过Metascape数据平台,进行GO功能及KEGG通路富集分析;最后使用AutoDock软件分子对接验证。结果:真武汤活性成分共59个,交集靶点55个。主要活性成分包括谷甾醇、β-谷甾醇、豆甾醇等,PPI分析出真武汤治疗眩晕的核心靶点可能为TNF、IL6、AKT1和CASP3等。富集分析主要涉及的通路有脂质和动脉粥样硬化通路、癌症、神经活性配体-受体相互作用通路等。分子对接证明核心成分与靶点结合活性良好。结果:真武汤治疗眩晕具有多成分、多靶点、多通路的特点,主要机制与促进血液循环、防止动脉粥样硬化、减轻炎症及凋亡反应、促进神经细胞再生等方面有关。Objective:To explore the potential mechanism of Zhenwu Decoction in treating vertigo by network pharmacology and molecular docking technology.Methods:The active ingredients and potential targets of Zhenwu Decoction were obtained through TCMSP and UniProt databases respectively;GeneCards,DRUGBANK and OMIM databases were used to obtain relevant targets of vertigo diseases;Venn diagrams were drawn and intersection targets were obtained,and PPI networks were generated through STRING database to screen key target sites and visualize them;GO function and KEGG pathway enrichment analysis were performed through Metascape data platform;and finally,molecular docking verification was performed using AutoDock software.Results:There were 59 active ingredients and 55 intersection targets in Zhenwu Decoction.The main active ingredients included sitosterol,β-sitosterol,stigmasterol,etc.PPI analysis showed that the core targets of Zhenwu Decoction in treating vertigo may be TNF,IL6,AKT1 and CASP3,etc.The pathways mainly involved in the enrichment analysis include lipid and atherosclerosis pathways,cancer,and neuroactive ligand-receptor interaction pathways.Molecular docking proved that the core components had good binding activity with the target.Results:Zhenwu decoction has the characteristics of multi-component,multi-target,and multi-pathway in the treatment of vertigo.The main mechanism is related to promoting blood circulation,preventing atherosclerosis,reducing inflammation and apoptosis,and promoting nerve cell regeneration.
