摘 要:通过响应面法优化纳豆激酶肠溶胶囊的制备工艺以提高胶囊的包埋率,并在人工胃肠液中验证纳豆激酶胶溶肠囊对纳豆激酶的保护作用。探讨了海藻酸钠浓度、羧甲基纤维素钠浓度和氯化钙浓度对纳豆激酶肠溶胶囊包埋率的影响。在单因素实验的基础上,进行响应面优化,得到的最优制备条件为:海藻酸钠36.43 g/L、羧甲基纤维素钠3.15 g/L、氯化钙7.69 g/L,包埋率为87.55%。将制得的纳豆激酶肠溶胶囊依次放于人工胃液和人工肠液中在37℃、180 r/min条件下分别处理2和3 h,同时将纳豆激酶裸酶放于人工胃液中相同条件下处理2 h作为对比。纳豆激酶肠溶胶囊在肠液中酶活为20.35 FU/mL,而裸酶酶活仅为1.36 FU/mL。模拟胃肠液实验结果表明制得的纳豆激酶肠溶胶囊具备显著的抵御胃酸的保护作用,为纳豆激酶的保健药品开发提供了理论依据和工艺参数。The preparation process of nattokinase enteric-coated capsules was optimized using response surface methodology to improve the encapsulation efficiency of the capsules,and the protective effect of nattokinase enteric-coated capsules on nattokinase was verified in artificial gastrointestinal fluid.The effects of sodium alginate concentration,sodium carboxymethylcellulose concentration and calcium chloride concentration on the encapsulation rate of nattokinase enteric-coated capsules were investigated.Based on the single-factor experiment,response surface optimization was performed,and the optimal preparation conditions were determied as follows:sodium alginate 36.43 g/L,sodium carboxymethylcellulose 3.15 g/L,calcium chloride 7.69 g/L,achieving an encapsulation rate of 87.55%.The prepared nattokinase entericcoated capsules were successively placed in artificial gastric juice and artificial intestinal juice and treated at 37℃and 180 r/min for 2 and 3 h,respectively.Meanwhile,the nattokinase in its free form was placed in artificial gastric juice under the same conditions for 2 h as a control.The enzyme activity of the nattokinase enteric-coated capsules in the intestinal juice was 20.35 FU/mL,while that of the free enzyme was only 1.36 FU/mL.The results of simulated gastrointestinal fluid experiments showed that the produced nattokinase enteric-coated capsules possessed significant protective effect against gastric acid,providing a theoretical basis and process parameters for the development of nattokinase-based health products.
