摘　　要：目的 研究常山酮(Halofuginone, HF)对人肝癌细胞MHCC-97H的影响。方法 采用MTT法检测HF对MHCC-97H细胞的抑制作用;通过体外克隆形成实验、划痕实验、transwell实验观察MHCC-97H细胞的增殖、迁移和侵袭。结果 HF能明显抑制MHCC-97H细胞的增殖,IC50为0.050±0.0065μmol·L^(-1);克隆形成实验显示,HF呈浓度依赖性地抑制MHCC-97H细胞的增殖,与对照组相比,加药组的细胞克隆团数目和大小明显减少;划痕及transwell实验表明,随着HF剂量的增大,细胞的相对迁移力显著下降,侵袭细胞数量显著下降。结论 HF对人高转移性肝癌细胞MHCC-97H具有良好的抵抗作用,可明显抑制MHCC-97H细胞的增殖、迁移及侵袭。Objective To study the influence of Halofuginone(HF)on human liver cancer cells MHCC-97H.Methods The effect of HF on the proliferation of MHCC-97H cells was detected with MTT assay.Clone formation in vitro,the scratch and transwell tests were used to study the proliferation,migration and invasion of cells.Results HF had significantly inhibitory effect on the proliferation of MHCC-97H cells,IC50 is 0.050±0.0065μmol‧L^(-1).It was found that HF inhibited the growth of MHCC-97H cells in a concentration-dependent manner.The number and size of cell clone clusters decreased significantly in the treated group as compared to the control group.The scratch and transwell tests demonstrated that the relative migration rate of HF decreased significantly and the number of invasive cells decreased markedly as the concentration of HF increased.Conclusion HF has good inhibitory activity on human highly metastatic liver cancer MHCC-97H cells and can significantly inhibit their proliferation,migration and invasion in vitro.
