摘　　要：目的 探讨正常宫颈、宫颈上皮内瘤变(CIN)和宫颈癌组织中细胞周期蛋白D1(Cyclin D1)和脆性组氨酸三联体(FHIT)的表达及意义。方法 采用qRT-PCR、Western blot和免疫组化染色法检测Cyclin D1和FHIT在15例正常宫颈组织、30例CIN组织和40例宫颈癌组织中的表达情况,对其与宫颈癌临床病理特征的关系进行分析。结果 与正常宫颈组织相比,CIN和宫颈癌组织中Cyclin D1的mRNA、蛋白表达水平及阳性表达率均显著升高(P<0.05);FHIT的mRNA、蛋白表达水平及阳性表达率均显著降低(P<0.05)。宫颈癌组织中,伴有低分化、淋巴结转移患者中Cyclin D1的阳性表达率高于中高分化、无淋巴结转移的患者(P<0.05);FHIT与年龄、临床分期、病理分级、病理分型及淋巴结转移无关(P>0.05);Cyclin D1与FHIT表达水平呈显著负相关(r=-0.086,P<0.05)。结论Cyclin D1过表达及FHIT低表达与宫颈癌发生发展密切相关。Objective To investigate the expression and clinical significances of fragile histidine triad(FHIT)and Cyclin D1 in normal cervical tissues,cervical intraepithelial neoplasia(CIN)tissues and cervical carcinoma tissues.Methods qRT-PCR,Western blot and immunohistochemistry was used to measure the expression of Cyclin D1 and FHIT in 15 cases of normal cervical tissues,30 cases of CIN tissues and 40 cases of cervical carcinoma tissues.Analyzed the correlation between Cyclin D1 and FHIT and clinicopathological factors of cervical carcinoma.Results Compared with the normal cervical tissues,the mRNA and protein expression level and positive expression rates of Cyclin D1 in CIN and cervical carcinoma tissues were significantly higher(P<0.05),the mRNA and protein expression level and positive expression rates of FHIT in CIN and cervical carcinoma tissues were significantly lower(P<0.05).In the cervical carcinoma tissues,the positive expression rates of Cyclin D1 which were in poorly differentiated or with lymph node metastasis were significantly higher than those in middle to high differentiated or without lymph node metastasis(P<0.05).The positive expression rates of FHIT were not related to age,clinical stage,pathological grade,pathological type and lymph node metastasis(P>0.05).The expression of Cyclin D1 was negatively correlated with FHIT(r=-0.086,P<0.05).Conclushion High expression of Cyclin D1 and low expression of FHIT are closely related to the occurrence and development of cervical carcinoma.
