摘　　要：目的:研究骨肉瘤组织及细胞中膜联蛋白A3(annexin A3,ANX A3)差异表达的意义。方法:通过免疫组化分析ANX A3在骨肉瘤组织及肉瘤旁组织表达水平的差异;使用Western blot法检测人骨肉瘤细胞株、成骨细胞株、ANX A3过表达或敲低骨肉瘤细胞株ANX A3表达水平差异;使用划痕实验和细胞计数试剂盒分析ANX A3过表达株或敲低骨肉瘤细胞株侵袭和增殖能力。结果:骨肉瘤组织中较肉瘤旁组织ANX A3表达阳性率显著升高,差异有统计学意义(P<0.05)。有远处转移病例的原发灶病理组织中ANX A3阳性率较无远处转移者明显升高,差异有统计学意义(P<0.05)。人骨肉瘤细胞株较成骨细胞株ANX A3表达水平显著升高2.96倍;与较成骨细胞株比,ANX A3过表达细胞株ANX A3水平明显上调,约为3.52倍,敲低株显著下调,差异均有统计学意义(均P<0.05);ANX A3过表达株迁移和增殖能力较敲低株显著增强,差异均有统计学意义(均P<0.05)。结论:ANX A3在骨肉瘤组织较瘤旁组织中显著表达,发生远处转移病例原发灶病理组织中ANX A3阳性率更高,ANX A3可促进骨肉瘤的增殖、迁移,可能成为骨肉瘤治疗的潜在靶点。Objective:To investigate the significance of the differential expression of annexin A3(ANX A3)in osteosarcoma tissues and cells.Methods:The expression of ANX A3 in osteosarcoma tissues and adjacent tissues was analyzed by immunohistochemistry,and the expression of ANX A3 in human osteosarcoma cell lines,osteoblast cell lines,ANX A3 overexpression or knockdown osteosarcoma cell lines was detected by Western blot.Scratch test and cell counting kit-8 were used to analyze the invasion and proliferation of ANX A3 overexpression or knockdown osteosarcoma cell lines.Results:The positive rate of ANX A3 expression in osteosarcoma tissues was significantly higher than that in adjacent tissues,and the difference was statistically significant(P<0.05).The positive rate of ANX A3 expression in pathological tissues of patients with distant metastasis was significantly higher than that of patients without distant metastasis,the difference was statistically significant(P<0.05).The expression level of ANX A3 in human osteosarcoma cell line was 2.96 times higher than that in osteoblast cell line,the difference was statistically significant(P<0.05).Compared with osteoblast cell lines,the level of ANX A3 in overexpressing cell lines was significantly up-regulated by about 3.52 times,while that in knockout cell lines was significantly down-regulated,and the difference was statistically significant(P<0.05).The migration and proliferation ability of ANX A3 overexpression strain was significantly higher than that of knockdown strain,the difference was statistically significant(P<0.05).Conclusions:Compared with adjacent tissues,ANX A3 is significantly expressed in osteosarcoma tissues,and the positive rate of ANX A3 is higher in pathological tissues of patients with distant metastasis.ANX A3 may promote the proliferation and migration of osteosarcoma,and may become a potential target for the treatment of osteosarcoma.
