摘  要:空间转录组学技术能够对生物样本的组织空间进行多维度研究,不但可以分析空间组织特征,还可以研究组织细胞中的基因表达与细胞通讯互作。肝癌是发病率和病死率最高的实体恶性肿瘤之一,其中耐药性和免疫抑制微环境是引发肝癌复发转移,导致肿瘤相关死亡的主要原因。近年来,空间转录组学技术在解析肝癌的遗传异质性、免疫微环境和细胞间通信互作等方面取得了许多重要研究成果,为基于新抗原靶点的药物研发和精准医疗提供了科学理论依据。本文旨在简述空间转录组学技术,并对其目前在肝癌微环境异质性和精准医疗中的应用进行综述,以期为肝癌的生物学机制探索和今后的防治提供一定的见解。Spatial transcriptomics technology can be used to study the tissue space of biological samples in many dimensions,not only to analyze the characteristics of spatial tissue,but also to study the gene expression and cell communication interaction in tissue cells.Hepatocellular carcinoma is one of the solid malignant tumors with the highest morbidity and mortality,in which drug resistance and immunosuppression microenvironment are the main causes of recurrence and metastasis of hepatocellular carcinoma,leading to tumor-related deaths.In recent years,spatial transcriptomics technology has made many important research achievements in analyzing the genetic heterogeneity,immune microenvironment and cell interaction communication of hepatocellular carcinoma,which provides scientific theoretical basis for drug development and precision medicine based on new antigen targets.In this paper,the spatial transcriptomics technology is briefly introduced,and its current application in the micro-environment heterogeneity and precision medicine of hepatocellular carcinoma is reviewed,so as to provide some insights for the biological mechanism exploration and future prevention and treatment of hepatocellular carcinoma.
