摘 要:目的建立1,1-二苯基-2-三硝基苯肼自由基(1,1-Diphenyl-2-picrylhydrazyl,DPPH)联合气相色谱和质谱联用技术(Gas Chromatography-Mass Spectrometry,GC-MS)快速筛选棒毛络新妇油脂中的抗氧化活性成分。方法采用DPPH自由基化学反应与GC-MS色谱分析相结合的方法进行研究,通过分析棒毛络新妇油脂与自由基反应前后色谱图的变化来筛选抗氧化成分,并以各化合物的色谱峰面积降低率评价其抗氧化活性。同时进行方法学考察,并采用酶标仪法测定对照品的自由基清除能力,以验证DPPH-GC-MS方法的抗氧化成分筛选结果。结果从棒毛络新妇油脂中共鉴定出22个化合物,占总量的89.67%,主要为脂肪酸和脂肪酸酯,其中油酸甲酯和亚油酸甲酯相对含量较高。通过DPPH-GC-MS方法从中快速筛选到16个抗氧化成分,其中2,4-二叔丁基苯酚、硬脂酸乙酯、亚油酸、亚油酸乙酯、油酸乙酯和棕榈酸乙酯的抗氧化能力较强。活性验证结果表明DPPH-GC-MS方法可靠,可对天然产物中的抗氧化活性物质进行快速筛选和鉴定。结论DPPH-GC-MS方法可简便高效地筛选出棒毛络新妇油脂中的抗氧化成分,为进一步开发利用棒毛络新妇资源提供实验依据。Objective To establish a gas chromatography mass spectrometry(GC-MS)technique using 1,1-Diphenyl-2-picrylhydrazyl(DPPH)as a free radical for rapid screening of antioxidant active components in the oil extracted from Bangmaoluoxinfu(Trichonephila clavata).Methods The method combining GC-MS chromatographic analysis with DPPH radical chemical reaction was used to screen antioxidant components by analyzing the changes in the chromatographic of the oil extracted from Bangmaoluoxinfu(Trichonephila clavata)before and after reaction with DPPH.Furthermore,the antioxidant activity of each compound was evaluated by the rate of reduction of the peak area of the chromatograms.At the same time,to verify the screening results of antioxidants by the DPPH-GC-MS method,a methodological study was carried out,and the free radical scavenging ability of the control product was determined by a microplate reader.Result A total of 22 compounds were identified from the oil extracted from Bangmaoluoxinfu(Trichonephila clavata),accounting for 89.67%of the total,mainly consisting of fatty acids and esters,with relatively high levels of methyl oleate and methyl linoleate.Sixteen antioxidants were rapidly screened from them by the DPPH-GC-MS method,among which 2,4-Di-tert-butylphenol,ethyl stearate,linoleic acid,ethyl linoleate,ethyl oleate and palmitic acid ethyl ester showed strong antioxidant capacity.The results of activity verification showed that the DPPH-GC-MS method was reliable and can be used for rapid screening and identification of antioxidants in natural products.Conclusion The DPPH-GC-MS method can simply and efficiently screen out the antioxidants in the oil extracted from Bangmaoluoxinfu(Trichonephila clavata),and provided the experimental basis for further development and utilization of Bangmaoluoxinfu(Trichonephila clavata)resources.
