摘 要:为探讨广西地方猪与商品猪肌内脂肪沉积能力的差异及相关基因的表达调控特征,本研究选取广西地方猪品种巴马香猪、环江香猪、桂中花猪、隆林猪、德保猪和外来品种三元杂商品猪为研究对象,利用索氏浸提法测定各品种猪背最长肌中IMF含量,通过实时荧光定量PCR方法检测脂肪沉积相关基因脂蛋白脂酶(Lipoprotein lipase.LPL)、激素敏感性脂肪酶(Hormone-sensitive triglyceride lipase,HSL)、脂肪酸转位酶(CD36)的表达水平。结果表明,巴马香猪、桂中花猪、隆林猪、环江香猪和德保猪的肌内脂肪含量均显著高于三元杂商品猪(P<0.05)。LPL、CD36和HSL基因在6个猪品种中均有不同程度的表达。LPL基因在隆林猪中的相对表达量最高,在巴马香猪中的表达量最低;CD36基因在环江香猪中的表达量最高、在巴马香猪中的表达量最低;HSL基因在桂中花猪、隆林猪和德保猪中的表达量均显著高于三元杂商品猪(P<0.05)。进一步显示出了LPL、CD36和HSL等3个基因与猪的脂肪沉积可能相关,为研究广西本地品种猪与外来品种猪在脂肪沉积候选基因的分子机制提供了基础。In order to explore the differences in intramuscular fat deposition ability between Guangxi local pigs and commercial pigs,as well as the expression regulation characteristics of related genes,this study selected Guangxi local breeds Bama xiang pigs,Huanjiang xiang pigs,Guizhong spotted pigs,Longlin pigs,Debao pigs and foreign breeds of ternary hybrid commercial pigs as research objects.The content of IMF in the longest muscle of each breed of pig was determined by Soxhlet extraction method,and the expression levels of fat deposition related genes lipoprotein lipase(LPL),hormone sensitive triglyceride lipase(HSL),and fatty acid transferase(CD36)were detected by real-time fluorescence quantitative PCR method.The results showed that the intramuscular fat content of Bama Xiang pigs,Guizhong spotted pigs,Longlin pigs,Huanjiang Xiang pigs,and Debao pigs were significanty higher than that of temnary hybrid commercial pigs(P<0.05).LPL,CD36,and HSL genes were expressed to varying degroes in all six varieties.The relative expression level of LPL gene was highest in Longlin pigs and lowest in Bama Xiang pigs;The expression level of CD36 gene was highest in Huanjiang xiang pigs and lowest in Bama xiang pigs;The expression level of HSL gene in Guizhong spotted pigs,Longlin pigs,and Debao pigs were significantly higher than that in ternary hybrid commercial pigs(P<0.05).Further evidence suggested that LPL,CD36,and HSL genes may be associated with fat deposition in pigs,provided a basis for the research of molecular mechanisms of candidate genes for fat deposition in local and foreign pig breeds in Guangxi.
