黄山玉兰叶绿体基因组特征及系统发育分析

摘  要:为明确黄山玉兰(Yulania cylindrica)叶绿体基因组结构特征及其与同属类群系统发育关系,采用高通量测序技术对黄山玉兰叶绿体全基因组进行测序和分析,基于木兰科15个叶绿体全基因组序列重建黄山玉兰及其近缘种系统发育树。结果表明:黄山玉兰叶绿体基因组全长为160080 bp,分为1个大单拷贝区(88119 bp)、1个小单拷贝区(18779 bp)和2个反向互补区(26591 bp);共注释得到113种基因,包括79种蛋白质编码基因、30种tRNA基因和4种rRNA基因,其中10种蛋白质编码基因和6种tRNA基因含有内含子;基于72个共享蛋白质编码基因序列的系统发育树显示黄山玉兰和玉兰(Y.denudata)的亲缘关系较近。研究结果对于进一步探讨广义木兰属进化与系统发育关系具有重要参考价值。In order to determine the structural features of the chloroplast genome of Yulania cylindrica and the phylogenetic relationship with the taxa of the same genus,high-throughput sequencing technology was adopted to sequence and analyze the complete chloroplast genome of Y.cylindrica in this study.Moreover,the phylogenetic trees of Y.cylindrica and its related species were reconstructed based on the 15 complete chloroplast genome sequences of Magnoliaceae.The results showed that the total genome size is 160080 bp in length,with one large single-copy(LSC,88119 bp)region and one small single-copy(SSC,18779 bp)region,separated by two copies of inverted repeats(IR,26591 bp)regions.The genome encodes a total of113 unique genes,including 79 protein-coding genes,30 t RNA genes,and 4 rRNA genes,and it includes 10protein-coding genes and 6 tRNA genes possess introns.The phylogenetic tree constructed based on 72shared protein-coding gene sequences indicated that Y.cylindrica has a close relationship with Y.denudata.Our findings were highly significant for further research on the evolution and phylogenetic relationships within the broad-sense Yulania genus.It was highly valuable as references for further exploring the evolutionary and phylogenetic relationships within the broad-sense Yulania genus.
