摘 要:目的探讨高氧液对顺铂化疗的乳腺癌耐顺铂细胞MCF7/DDP(Michigan Cancer Foundation-7)种植瘤裸鼠瘤体体积、缺氧诱导因子-1α(hypoxia inclucible factor-1α,HIF-1α)和多药耐药基因-1(multidrug resistanoe gene-1,MDR-1)表达的影响。方法MCF7/DDP细胞接种于4周雌性裸鼠,喂养6周后根据体重分别选取5只裸鼠作为A、B组,两组均给予顺铂化疗(剂量6.25 mg/kg),连续化疗1周后按照20 mg/kg B组予以生理盐水静脉推注,A组静脉推注高氧液,采集裸鼠0.2 mL动脉血,行动脉血气分析,并以RT-PCR检测HIF-1α、MDR-1表达水平;再处死裸鼠并分离肿瘤组织,测量组织直径。结果A组裸鼠pH值、PO2均高于B组,差异有统计学意义(P<0.05)。A组裸鼠HIF-1α、MDR-1相对表达量和肿瘤体积、质量均低于B组,差异有统计学意义(P<0.05)。结论种植瘤裸鼠经高氧液输注可能有效改善缺氧状态,同时下调HIF-1α及MDR-1表达,增强化疗敏感性。Objective To investigate the effect of hyperoxygenated solution on the tumor volume,the expression of hypoxia-inducible factor-1α(HIF-1α),and multidrug resistance gene-1(MDR-1)in MCF7/DDP(Michigan Cancer Foundation-7)DDP-resistant breast cancer cells implanted in nude mice after chemotherapy.Method The MCF7/DDP cells were inoculated into 4-week-old female nude mice.After feeding for 6 weeks,five nude mice were selected according to their body weight as Group A and Group B.Both groups were given cisplatin chemotherapy(dose 6.25 mg/kg).After one week of continuous chemotherapy,Group B was administered 20 mg/kg of physiological saline via intravenous injection,while Group A was given hyperoxygenated solutions intravenously.Samples of 0.2 mL of arterial blood were collected from the nude mice for arterial blood gas analysis;the expression levels of HIF-1αand MDR-1 were assessed using RT-PCR,the nude mice were then executed,and tumor tissues were isolated for the measurement of tumor diameter.Result The pH value and PO 2 were significantly higher in Group A compared to Group B,with statistically significant differences(P<0.05).Moreover,the relative expression levels of HIF-1αand MDR-1,as well as tumor volume and size in Group A,were significantly lower than those in Group B,which was also statistically significant(P<0.05).Conclusion Hyperoxygenated solution infusion effectively promotes blood circulation and alleviates hypoxia in tumor-bearing nude mice while downregulating HIF-1αand MDR-1 expression levels and enhancing chemotherapy sensitivity.
