摘　　要：目的探讨SRPX2(含sushi重复蛋白X连锁2)在胃癌组织中的表达、免疫细胞浸润以及预后的关系,以期进一步明确SRPX2在胃癌中的生物学功能及预后价值,为今后实施个体化预防及诊治提供线索和参考。方法使用多个数据库分析SRPX2基因在泛癌及胃癌中的表达,使用临床组织样本进行验证并与临床特征相关联。通过Kaplan Meier-Plotter数据库和R语言分析SRPX2与患者总体生存期(OS)、疾病特异性生存期(DSS)以及无进展间隔期(PFI)之间的关系。使用STRING数据库和GEPIA2数据库对SRPX2共表达基因进行PPI分析和GO/KEGG功能富集。TIMER数据库分析胃癌组织中SRPX2表达与免疫细胞浸润之间的联系。结果基于TCGA和GEPIA数据库分析提示,SRPX2在胃癌组织中相对高表达,并且与临床病理特征相关。SRPX2可能是胃癌的独立预后因素,并且该基因的高表达与胃癌不良预后相关。GO/KEGG功能富集分析提示,SRPX2与“局灶粘连”、“蛋白多糖”和“蛋白质的消化和吸收”等通路相关。TIMER数据库分析显示,SRPX2的表达与多种免疫细胞的浸润相关。结论SRPX2在胃癌组织中表达上调并与胃癌患者肿瘤细胞免疫浸润及不良预后相关,有可能成为胃癌患者治疗以及判断预后的分子靶点。Objective To investigate the relationship among the expression of SRPX2(including sushi repeat protein X-linked 2)in gastric cancer tissues,immune cell infiltration and prognosis,in order to further clarify the biological function and prognostic value of SRPX2 in gastric cancer,and provide clues and references for future individualized prevention,diagnosis and treatment.Methods Expression of the SRPX2 gene in pan-and gastric cancers was analyzed using multiple databases,validated using clinical tissue samples and correlated with clinical features.Relationships between SRPX2 and patient overall survival(OS),disease-specific survival(DSS),and progression-free interval(PFI)were analyzed by the Kaplan Meier-Plotter database and R language.PPI analysis and GO/KEGG functional enrichment of SRPX2 co-expressed genes were performed using the STRING database and the GEPIA2 database.The TIMER database was used to analyze the association between SRPX2 expression and immune cell infiltration in gastric cancer tissues.Results Analysis based on TCGA and GEPIA databases suggested that SRPX2 was relatively highly expressed in gastric cancer tissues,which is closely associated with clinicopathologic features.SRPX2 may be an independent prognostic factor for gastric cancer,and high expression of this gene is associated with poor prognosis in gastric cancer.GO/KEGG functional enrichment analysis suggested that SRPX2 was closely related to the“focal adhesion”,“proteoglycans in cancer”and“protein digestion and absorption”pathways.TIMER database analysis showed that SRPX2 expression was correlated with the infiltration of various immune cells.Conclusions SRPX2 expression is up-regulated in gastric cancer tissues and correlates with immune infiltration of tumor cells and poor prognosis in gastric cancer patients,which could potentially be a molecular target for the treatment of gastric cancer patients as well as for determining their prognosis.
