摘 要:目的:研究Lin28基因在肝母细胞瘤(HB)中的表达情况,并探讨Lin28的表达对HB细胞生物学功能的影响。方法:综合分析癌症基因组图谱(TCGA)数据库和3个不同来源的基因表达数据库(GEO)数据集中Lin28基因的表达差异情况;通过转染构建的siRNA分别敲低HB细胞系Huh-6和HepG2的表达水平,通过细胞克隆集落形成实验、CCK-8、流式细胞术检测Lin28对HB细胞生物学表型的影响。结果:在TCGA数据库中,Lin28在大多数肿瘤中的表达均上调(P<0.05)。在GEO数据库中,Lin28在HB肿瘤组织较癌旁组织表达上调(P<0.05)。成功构建转染si-Lin28后,Lin28在mRNA及蛋白水平表达显著下降(P<0.05);敲低Lin28的表达可以显著抑制HB细胞系Huh-6和HepG2增殖能力(P<0.01),增加细胞凋亡率(P<0.05)。结论:Lin28在HB中表达上调,敲低Lin28的表达可以抑制细胞恶性表型,提示Lin28可能是HB一个潜在的治疗靶点。Objective:To investigate the expression of Lin28 gene in hepatoblastoma(HB)and explore its impact on biological phenotypes.Methods:The expression of Lin28 in pan-cancer and its expression in hepatoblastoma were analyzed using data from The Cancer Genome Atlas(TCGA)and three independent Gene Expression Omnibus(GEO)datasets.siRNA-mediated knockdown of Lin28 was performed in HB cell lines Huh-6 and HepG2.Small interfering RNAs(siRNAs)targeting Lin28 genes were constructed to transfect Huh-6 and HepG2 cells,and the interfering effect was verified at mRNA and protein levels.The effect of Lin28 on the biological phenotypes of HB cells was assessed by colony formation assays,CCK-8 assays,and flow cytometry.Results:In the TCGA database,Lin28 was found to be upregulated in the majority of tumors(P<0.05).In the GEO datasets,Lin28 expression was significantly higher in HB tumor tissues than adjacent non-tumor tissues(P<0.05).Successful transfection of si-Lin28 led to a significant reduction in Lin28 expression at both mRNA and protein levels(P<0.05).Knockdown of Lin28 markedly inhibited the proliferation of HB cell lines Huh-6 and HepG2(P<0.01)and increased the apoptosis rate(P<0.05).Conclusion:Lin28 is upregulated in HB,and its knockdown suppresses the malignant phenotype of HB cells,suggesting that Lin28 may serve as a potential therapeutic target for HB.
