银杏内酯B介导IKK/NF-κB通路对肺腺癌A549细胞增殖及凋亡的调控作用

摘  要:为探究银杏内酯B对人肺腺癌(A549)细胞增殖和凋亡的影响及IκB激酶/核因子κB(IKK/NF-κB)信号通路的调控作用,体外培养A549细胞,将其分为对照组(相同体积二甲基亚砜溶剂)和不同浓度(4、8、16、32μmol/L)银杏内酯B组,干预24 h,细胞计数试剂盒-8(CCK-8)检测细胞活力筛选最适浓度的银杏内酯B用于后续试验。随后将A549细胞分为对照组、银杏内酯B组(16μmol/L银杏内酯B)、阳性药物组(0.4 mg/L阿霉素)、抑制剂组(16μmol/L银杏内酯B+10μmol/L IKK/NF-κB通路抑制剂BAY11-7082)和激活剂组(16μmol/L银杏内酯B+1μmol/L IKK/NF-κB通路激活剂Prostratin),干预24 h。5-乙炔基-2′脱氧尿嘧啶核苷(EdU)测定细胞增殖能力;Hoechst 33258染色试剂盒测定细胞凋亡能力;蛋白免疫印迹法检测细胞周期蛋白D1(CyclinD1)、半胱氨酸蛋白酶-3(Caspase-3)及IKK/NF-κB通路相关蛋白表达水平。根据CCK-8试验结果,选择16μmol/L银杏内酯B用于后续试验。与对照组比较,银杏内酯B组和阳性药物组A549细胞增殖率、CyclinD1、p-NF-κB p65/NF-κB p65和p-IκBα/IκBα比值显著降低(P<0.05),凋亡率和Caspase-3蛋白表达水平显著升高(P<0.05);与银杏内酯B组比较,抑制剂组细胞增殖率、CyclinD1、p-NF-κB p65/NF-κB p65和p-IκBα/IκBα比值显著降低(P <0.05),凋亡率和Caspase-3蛋白表达水平显著升高(P <0.05),激活剂组细胞增殖率、CyclinD1、p-NF-κB p65/NF-κB p65和p-IκBα/IκBα比值显著升高(P<0.05),凋亡率和Caspase-3蛋白表达水平显著降低(P<0.05)。银杏内酯B能通过调控IKK/NF-κB通路信号转导抑制A549细胞的增殖,并诱导其凋亡。To investigate the effect of ginkgolide B on the proliferation and apoptosis of human lung adenocarcinoma(A549)cells and the regulatory effect of IκB kinase/nuclear factorκB(IKK/NF-κB)signaling pathway.A549 cells were cultured in vitro and divided into control group(same volume DMSO solvent)and different concentrations(4,8,16,32μmol/L)of ginkgolide B group.After 24 hours of intervention,cell counting kit-8(CCK-8)was used to detect cell viability to screen the optimal concentration of ginkgolide B for subsequent experiments.Then A549 cells were divided into control group,ginkgolide B group(16μmol/L ginkgolide B),positive drug group(0.4 mg/L adriamycin),inhibitor group(16μmol/L ginkgolide B+10μmol/L IKK/NF-κB pathway inhibitor BAY11-7082)and activator group(16μmol/L ginkgolide B+1μmol/L IKK/NF-κB pathway activator Prostratin),intervention for 24 hours.5-ethynyl-2′-deoxyuridine(EdU)was used to measure cell proliferation.Hoechst 33258 staining kit was used to detect cell apoptosis.Western blot(WB)was used to detect the expression levels of CyclinD1,Caspase-3 and IKK/NF-κB pathway-related proteins.According to the results of CCK-8 experiment,16μmol/L ginkgolide B was selected for subsequent experiments.Compared with the control group,the proliferation rate of A549 cells and the protein expression levels of CyclinD1,p-NF-κB p65/NF-κB p65 and p-IκBα/IκBαin the ginkgolide B group and the positive drug group significantly decreased(P<0.05),and the apoptosis rate and Caspase-3 protein expression level significantly increased(P<0.05).Compared with ginkgolide B group,cell proliferation rate,CyclinD1,p-NF-κB p65/NF-κB p65 and p-IκBα/IκBαratio significantly decreased in inhibitor group(P<0.05),while apoptosis rate and Caspase-3 protein expression level significantly increased(P<0.05).In the activator group,cell proliferation rate,CyclinD1,p-NF-κB p65/NF-κB p65 and p-IκBα/IκBαratios significantly increased(P<0.05),while apoptosis rate and Caspase-3 protein expression levels significantly decreas
