摘  要:目的探讨天花粉蛋白(TCS)对胃癌细胞生物活性及Livin基因mRNA水平的影响。方法将人类低分化胃癌细胞株BGC-823分为对照组和干预组,对照组无药物处理,干预组经过7、14和28μmol/L浓度的TCS处理,显微镜观察BGC-823细胞形态,MTT观察活力,流式细胞仪观察凋亡率,免疫印迹和qRT-PCR检测Livin蛋白和mRNA水平。结果7、14和28μmol/L天花粉蛋白使BGC-823细胞株形态逐渐变形皱缩,透明度降低,生长能力减慢,体积变大,细胞碎片漂浮,细胞活力随着TCS剂量升高而降低,呈现时间依赖性,BGC-823细胞凋亡率均高于对照组(P<0.05),Livin蛋白和mRNA水平低于对照组。结论天花粉蛋白抑制胃癌细胞BGC-823增殖,加快凋亡,与抑制Livin基因水平相关。Objective To explore the effect of Trichosanthin(TCS)on the biological activity of gastric cancer cells and the level of Livin gene.Methods Human poorly differentiated gastric cancer cell line BGC-823 was divided into control group and intervention group.The morphology of BGC-823 cells was observed under microscope,cell vitality was studied by MTT,apoptosis by flow cytometry,immunoblotting and qRT-PCR were used to determine Livin protein and mRNA.Results 7,14 and 28μmol/L TCS resulted in BGC-823 cells shape gradually deformed and shrunk,reduced transparency,slowed growth ability,decreased cell viability in dose dependent manner,the increased apoptosis rate of BGC-823 cells,the lower expression levels of Livin protein and mRNA in BGC-823 cells(P<0.05).Conclusion TCS inhibits the proliferation of human poorly differentiated gastric cancer BGC-823 cells and accelerates the cell apoptosis,which is related to the inhibition of Livin gene.
