摘 要:目的探究FGF21介导铁死亡对脑缺血再灌注大鼠神经损伤的改善作用及机制。方法SD大鼠随机分为假手术组(Sham组)、脑缺血再灌注组(I/R组)和脑缺血再灌注+FGF21治疗组(I/R+FGF21组),每组10只。采用线栓法建立MCAO/R模型;采用Zea Longa评分标准进行神经功能缺损评分;TCC染色检测大鼠脑梗死面积;HE染色观察大鼠脑组织病理损伤;组织铁含量试剂盒检测脑组织铁沉积;Western blot检测大鼠脑组织中TF、TFR、GPX4、FGFR1和NRG1蛋白表达情况。结果与I/R组相比,给予FGF21能降低脑缺血再灌注大鼠神经功能缺损评分,减少大鼠脑梗死面积并改善大鼠脑组织病理损伤,降低大鼠脑组织中铁含量以及TF和TFR蛋白表达并增加脑组织中GPX4蛋白表达,上调大鼠脑组织中FGFR1和NRG1蛋白表达。结论FGF21通过激活FGFR1/NRG1信号通路抑制铁死亡减少大鼠脑梗死面积并改善大鼠脑组织病理损伤。Objective To explore the improvement effect and mechanism of FGF21 mediated iron death on nerve injury in rats with cerebral ischemia-reperfusion.Methods SD rats were randomly divided into sham operation group(Sham group),cerebral ischemia-reperfusion group(I/R group)and cerebral ischemia-reperfusion+FGF21 treatment group(I/R+FGF21 group),with 10 rats in each group.The MCAO/R model was established by the thread-bolt method.Neurological impairment was evaluated by using Zea Longa score.The infarct size of rats was detected by TCC staining.The pathological injury in brain of rats was observed by HE staining.Iron deposition in brain tissue was detected by tissue iron content kit.The expressions of TF,TFR,GPX4,FGFR1 and NRG1 in rat brain were detected by Western blot.Results Compared with the I/R group,the administration of FGF21 can reduce the neural function deficit score,reduce the cerebral infarction size and improve the pathological injury in the brain tissue of rats,reduce the iron content and the expression of TF and TFR protein in the brain tissue of rats,increase the expression of GPX4 in the brain tissue of rats,and up-regulate the expression of FGFR1 and NRG1 protein in the brain tissue of rats.Conclusion FGF21 inhibits iron death by activating FGFR1/NRG1 signaling pathway to reduce cerebral infarction size and improve pathological brain injury in rats.
