摘  要:目的探讨补充适量短链脂肪酸对肠道屏障系统和炎性反应的调节作用。方法将30只小鼠随机分为对照组、模型组和模型组+SCFAs,每组10只,记录各组小鼠体重,运用气相色谱仪检测小鼠粪便样本中乙酸、丙酸和丁酸的含量。HE染色观察小鼠肠组织形态学变化;Western blot实验检测小鼠肠组织中NLRP6、Occludin和ZO-1蛋白表达;ELISA实验检测小鼠肠道组织中的炎性因子白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)和白细胞介素6(IL-6)的表达。结果与对照组相比,模型组小鼠的体重明显降低(P<0.05)。SCFAs治疗后,模型组小鼠粪便中乙酸、丙酸和丁酸的含量明显增加(P<0.05);小鼠肠道组织NLRP6、Occludin和ZO-1蛋白表达增加(P<0.05);小鼠肠道组织IL-1β、IL-6和TNF-α表达降低(P<0.05)。结论补充适量短链脂肪酸能降低肠道炎性反应,增强肠道黏膜屏障功能。Objective To explore the regulation of intestinal barrier system and inflammatory response by supplementing proper amount of short-chain fatty acids.Methods Thirty mice were randomly divided into control group,model group and model group+SCFAs,with 10 mice in each group.The weight of mice in each group was recorded,and the contents of acetic acid,propionic acid and butyric acid in stool samples of mice were detected by gas chromatograph.The changes of intestinal morphology were observed by HE staining.The expressions of NLRP6,Occludin and ZO-1 in intestinal tissue of mice were detected by Western blot.The expressions of interleukin-1β(IL-1β),tumor necrosis factorα(TNF-α)and interleukin-6(IL-6)in intestinal tissues of mice were detected by ELISA.Results Compared with the control group,the body weight of mice in the model group was significantly decreased(P<0.05).After SCFAs treatment,the contents of acetic acid,propionic acid and butyric acid in feces of model group were significantly increased(P<0.05).The expressions of NLRP6,Occludin and ZO-1 in intestinal tissue of mice were increased(P<0.05).The expressions of IL-1β,IL-6 and TNF-α in intestinal tissue of mice were decreased(P<0.05).Conclusions Supplementation of appropriate amount of short-chain fatty acids can reduce intestinal inflammatory response and enhance intestinal mucosal barrier function.
