土木香内酯对人骨肉瘤MG63细胞增殖、凋亡、侵袭与迁移的影响

摘  要:目的探讨土木香内酯对人骨肉瘤MG63细胞增殖、凋亡、侵袭与迁移的影响及可能机制。方法体外培养人骨肉瘤MG63细胞,取对数期生长的MG63细胞,分为空白对照组和土木香内酯(浓度分别为2.5、5.0、10.0μmol/L)组。MTT实验检测MG63细胞的增殖能力;流式细胞仪检测MG63细胞的凋亡率;Transwell实验检测MG63细胞的侵袭与迁移能力;Western blot实验检测MG63细胞PDK1、Akt与GSK3β蛋白的表达。结果与对照组相比,2.5、5.0、10.0μmol/L的土木香内酯溶液处理后,MG63细胞的增殖、侵袭与迁移能力明显降低,凋亡率明显升高,且呈浓度依赖性(P<0.05)。与空白对照组相比,2.5、5.0、10.0μmol/L土木香内酯组MG63细胞p-PDK1、p-Akt和p-GSK3β蛋白的表达均显著降低(P<0.05)。结论土木香内酯通过抑制PDK1/Akt/GSK3β信号通路抑制MG63细胞的增殖、侵袭与迁移能力,促进MG63细胞凋亡。Objective To investigate the effects and possible mechanisms of alantolactone on proliferation,apoptosis,invasion and migration of human osteosarcoma MG63 cells.Methods Human osteosarcoma MG63 cells were cultured in vitro,and MG63 cells with logarithmic growth were divided into blank control group and alantolactone(concentration 2.5,5.0,10.0μmol/L,respectively)group.The proliferation ability of MG63 cells was detected by MTT assay.The apoptosis rate of MG63 cells was detected by flow cytometry.The invasion and migration ability of MG63 cells were detected by Transwell assay.The expressions of PDK1,Akt,GSK3βin MG63 cells were detected by Western blot assay.Results Compared with the control group,the proliferation,invasion and migration ability of MG63 cells were significantly decreased and the apoptosis rate was significantly increased in a concentration-dependent manner after being treated with 2.5,5.0 and 10.0μmol/L alantolactone solution(P<0.05).Compared with blank control group,the expressions of P-PDK1,p-Akt and P-GSK3βin MG63 cells in 2.5,5.0 and 10.0μmol/L alantolactone groups were significantly decreased(P<0.05).Conclusions Alantolactone can inhibit the proliferation,invasion and migration of MG63 cells,and promote the apoptosis of MG63 cells by inhibiting PDK1/Akt/GSK3β signaling pathway.
