摘 要:目的探讨酸、脱氧胆酸(DCA)对BAR-T细胞SOX2表达的影响,分析SOX2与Barrett食管肠化生发展食管腺癌的相关性。方法通过体外培养人BAR-T细胞,用不同浓度酸pH=3、4、5,脱氧胆酸100μmol/L、200μmol/L、400μmol/L培养液干预,于6 h、12 h、24 h后应用MTT方法检测各组BAR-T细胞增殖;于12 h后应用Western blot和qRT-PCR方法观察各组细胞SOX2蛋白及mRNA表达水平的变化。结果DCA=100μmol/L组6 h时细胞增殖比升高,12 h、24 h时细胞增殖比降低;pH=3、4、5,DCA=200μmol/L、400umol/L组随着干预时间延长、酸和脱氧胆酸浓度增大,细胞增殖比降低,凋亡率明显升高(P<0.05)。不同浓度酸、脱氧胆酸组细胞中SOX2蛋白及mRNA的表达较正常对照组下降,且随着酸、脱氧胆酸浓度升高,SOX2蛋白及mRNA水平均呈现逐渐降低(P<0.05)。结论高浓度酸、脱氧胆酸抑制食管细胞增殖,并使BAR-T细胞中SOX2蛋白及mRNA水平下调,与浓度呈负相关,可促进食管细胞肠上皮化生,从而导致食管腺癌的发生。Objective To investigate the effects of acid and deoxycholic acid(DCA)on SOX2 expression in BAR-T cells and analyze the correlation between SOX2 and Barrett′s intestinal metaplasia and development of esophageal adenocarcinoma.Methods Human BAR-T cells were cultured in vitro with different concentrations of acid pH=3,4,5,DCA 100μmol/L,200μmol/L,400μmol/L culture solution.The proliferation of BAR-T cells in each group was detected by MTT method after 6 h,12 h and 24 h.The expression levels of SOX2 protein and mRNA in each group after 12 h were detected by Western blot and qRT-PCR.Results The cell proliferation ratio was increased at 6 h and decreased at 12 h and 24 h in the 100μmol/L DCA group.In pH=3,4,5,DCA=200μmol/L,400μmol/L groups,with the extension of intervention time and the increase of acid and deoxycholic acid concentrations,the cell proliferation ratio was decreased and the apoptosis rate was increased significantly(P<0.05).The expression of SOX2 protein and mRNA in groups with different concentrations of acid and deoxycholic acid was decreased compared with the normal control group,and the levels of SOX2 protein and mRNA was decreased gradually with the increase of acid and deoxycholic acid concentration(P<0.05).Conclusions High concentration of acid and deoxycholic acid can inhibite the proliferation of esophageal cells and reduce the level of SOX2 protein and mRNA in BAR-T cells,which is negatively correlated with the concentration,and may promote intestinal metaplasia of esophageal cells,thus leading to the occurrence of esophageal adenocarcinoma.
