摘  要:探究不同森林地下水微生物群落组成和多样性及环境驱动因素,为热带森林研究提供基础数据。本研究利用宏基因组测序技术分析了海南岛热带雨林和橡胶林2种森林的地下水微生物群落组成、多样性以及环境因素。结果表明:1)热带雨林与橡胶林地下水微生物群落构成存在明显差异,古菌群落方面,橡胶林奇古菌门(Thaumarchaeota)占比极显著高于热带雨林(P<0.001),而广古菌门(Euryarchaeota)则显著低于热带雨林(P<0.05);细菌群落中,橡胶林变形菌门(Proteobacteria)略高于热带雨林,而拟杆菌门(Bacteroidetes)则低于热带雨林(P<0.05);真菌群落中橡胶林子囊菌门(Ascomycota)显著高于热带雨林(P<0.05),而毛霉菌门(Mucoromycota)极显著低于热带雨林(P<0.001)。2)热带雨林的地下水微生物α多样性高于橡胶林,从热带雨林到橡胶林古菌群落的α多样性极显著降低(P<0.001),细菌群落的α多样性显著降低(P<0.05),真菌群落的α多样性显著升高(P<0.05)。主坐标分析(PCoA)和相似性分析揭示,海南岛橡胶林和热带雨林地下水微生物β多样性具有极显著差异(古菌:R=0.666,P=0.001;细菌:R=0.519,P=0.001;真菌:R=0.384,P=0.001)。3)地下水的溶解性总固体、海拔和氧化还原电位分别是影响古菌群落、细菌群落和真菌群落最重要的环境因子,大气压、重碳酸盐和海拔对地下水微生物β多样性具有显著影响,其中海拔是最主要的环境因素。The objective of this study was to explore the composition and diversity of groundwater microbial communities and environmental driving factors in different forests,so as to provide basic data for the study of tropical forests.Metagenomic sequencing technology was used to analyze the composition and diversity of groundwater microbial communities and environmental factors in two types of forests,tropical rainforest and rubber forest occurring in Hainan Island.The results showed that 1)there were obvious differences in the composition of groundwater microbial communities between tropical rainforest and rubber forest(P<0.001),and the proportion of archaeal community was higher than that of tropical rainforest,while that of Euryarchaeota was significantly lower than that of tropical rainforest(P<0.05).Among the bacterial communities,Proteobacteria was slightly higher in rubber forest than in tropical rainforest,while Bacteroidetes was lower than that in tropical rainforest(P<0.05).In the fungal community,Ascomycota was significantly higher than that in tropical rainforest(P<0.05),while Mucoromycota was significantly lower than in tropical rainforest(P<0.001).2)Theαdiversity of groundwater microorganisms in tropical rainforests was higher than that in rubber forests.Theαdiversity of archaeal communities from tropical rainforests to rubber forests significantly reduced(P<0.001),theαdiversity of bacterial communities significantly reduced(P<0.05),and theαdiversity of fungal communities significantly increased(P<0.05).Principal coordinate analysis(PCoA)and similarity analysis revealed theβdiversity of groundwater microorganisms,significant differences were observed between rubber forests and tropical rainforests in Hainan Island(archaea:R=0.666,P=0.001;bacteria:R=0.519,P=0.001;fungi:R=0.384,P=0.001).3)The total dissolved solids,altitude,and redox potential of groundwater were the most important environmental factors affecting archaeal,bacterial,and fungal communities,respectively.Atmospheric pressure,bicarbonate,and
