摘  要:目的:中低强度的运动会对宿主肠道菌群产生有益影响,但高强度或力竭性运动对肠道菌群影响的相关研究较少。本研究观察力竭运动对大鼠肠道菌群结构的影响,进一步理解力竭运动对肠道菌群的影响机制。方法:12只7周龄SD大鼠分为力竭组和对照组,力竭组采用递增运动强度的力竭运动为干预方式对大鼠进行为期6周的干预,对照组允许笼中自由活动。6周运动干预结束后24h内收集大鼠新鲜粪便样品,提取粪便细菌DNA,16srDNA高通量测序对大鼠肠道菌群进行分析。结果:Alpha多样性分析显示:对照组和力竭组肠道菌群的丰富度存在显著差异(P<0.05)。两组样本的肠道菌群多样性差异不具有统计学意义(P>0.05)。两组样本的菌群在门和纲水平上差异不显著,在目、科、属水平上存在显著差异的菌种,力竭组健康菌种如嗅杆菌科(Odoribacteraceae)、杆菌属(Odoribacter)、理研菌科(Rikenellaceae)、双歧杆菌(Bifidobacterium)等相对含量显著低于普通组,而一些有害菌如葡萄球菌科(Staphylococcaceae)等相对含量显著高于普通组(P<0.05)。结论:力竭性运动会改变大鼠肠道菌群的丰度,也会使部分健康菌种含量降低;部分有害菌种含量增加。Objective:Low and medium intensity exercise will have a beneficial impact on the host gut microbiota,but there are few studies on the effects of high intensity or exhaustive exercise on the gut microbiota.This study observed the effect of exhaustive exercise on gut microbiota structure of rats,which is conducive to further understanding the mechanism of exhaustive exercise on intestinal microflora.Methods:twelve SD rats aged 7 weeks were divided into exhaustion group and control group.Exhaustion exercise with increasing exercise intensity was used as intervention mode for 6 weeks.The control group was allowed to move freely in cage.Fresh fecal samples were collected within 24 hours after the 6-week exercise intervention,and fecal bacterial DNA was extracted,and the gut microbiota of rats was analyzed by 16srDNA high-throughput sequencing.Results:Alpha diversity analysis showed that there were significant differences in the richness of gut microbiota between the control group and the exhaustive exercise group(P<0.05).There was no statistical significance in the diversity of gut microbiota between the two groups(P>0.05).There were no significant differences in phylum and class level between the two groups of samples,while there were significant differences in order,family and genus level.The relative content of healthy bacteria such as Odoribacteraceae,Odoribacter,Rikenellaceae and Bifidobacterium in the exhausted group was significantly lower than that in the normal group.The relative content of some harmful bacteria,such as Staphylococcaceae,was significantly higher than that of normal group(P<0.05).Conclusion:Exhaustive exercise changed the abundance of gut microbiota in rats,and also reduced the content of some healthy bacteria.The content of some harmful bacteria increased.
